Die Untersuchungen befassen sich rnit dem Einsatz technischer Priparate von Starkesulfaten und Xunthor,ionus-Polysacchariden als Fallungsmittel fur Albumine. Wesentliche EinfluDgroRen auf die Fillungsquote sind das Konzentrationsverhaltnis von Proteinen zu Komplexbildnern sowie der pH-Wert und der Elektrolytgehalt des Extraktes. Die Fallung ist daneben vom Veresterungsgrad der Starkesulfate bzw. dem Molekulargewicht der Xanfhomonus-Polysaccharide abhangig. Albumin-Komplexe rnit Starkesulfaten unterscheiden sich von denen der Xanrhomonas-Polysaccharide durch eine hohere Stabilitat.
Erucasäurearme Zuchtstämme von Rapssamen (Einfach‐ oder Null‐Qualitätsraps) verdrängen auf Grund ihres höheren Gebrauchswertes traditionelle Sorten in zunehmendem Maße [1]. Auch in der DDR stehen inzwischen mit den Varietäten Marinus, Bellinda und Malux derartige Neuzüchtungen für eine agrarwirtschaftliche Nutzung zur Verfügung. Diese unterscheiden sich in ihrem Glucosinolatgehalt und ‐spektrum nur geringfügig von Standard‐Qualitätsraps, weisen jedoch zum Teil differenzierte Spaltungsmechanismen auf, die sich auf ihr Extraktionsverhalten bei der Fett‐ oder Proteingewinnung auswirken können [2]. Das folgt aus vergleichenden Untersuchungen an verschiedenen Rapssamen‐Varietäten, bei denen auch auf Unterschiede in Proteinmuster und der Morphologie hingewiesen wurde [3, 4]. Somit bestand Veranlassung, das Extraktionsverhalten wertbestimmender Sameninhaltsstoffe des Stammes Marinus, speziell der Glucosinolate und ihrer Autolyseprodukte sowie der Rohprodukte und sonstiger löslicher Verbindungen näher zu erkunden.
Die Effektivitat der extraktiven Eliminierung antinutritiver oder toxischer Bestandteile von Rapssamen, speziell Glucosinolate und deren Spaltprodukte, wie auch das AusmaS an Extraktionsverlusten wertbestimmender Inhaltsstoffe bei der Konzentratgewinnung hangt von der thermischen Vorbehandlung der Rohstoffe, den Extraktionsbedingungen und der morphologischen Beschaffenheit der Rohstoffe ab. Bei Extraktion von Samen oder Mehlen unterschiedlicher Rapsvarietaten mit Wasser bei pH 4,5 oder waSrigem Ethanol bei pH 6,O werden nur graduelle Unterschiede in der Loslichkeit von Stickstoffverbindungen und Kohlenhydraten festgestellt. Im Gegensatz d a m differiert die Loslichkeit von Glucosinolaten erheblich, was bei der Diffusionsextraktion intakter Samen mit Doppelqualitat besonders markant ist.
Membrantrennprozesse spielen seit l-erem in der Milch-und Fruchtsaftindustrie eine Rolle (1, 2). Sie sind aber auch im Hinblick auf die Gewinnung von Proteinisolaten aus pflanzlichen Rohstoffen (3 -5), u. a. Rapssamen mit Standard-Qualittit, untersucht worden, wobei verschiedenartige Extraktions-~t t e l zum Einsatz gelangten (6 -9 ) . In Ab-gkeit von den gewtihlten Milieubedingungen, speziell dern pH-Wert und dem Elektrolytgehalt, sowie verwendeten Zusatzstoffen, wie Polyvinylpyrrolidon, werden danach Endprodukte mit niedrigen Gehalten an Phytinsaure, Phenolsauren oder Glucosinolaten erhdten. Darilber hinaus sind durch selektive Extraktion in Kombination mit einer Ultra-und Diafiltration schadstoffme Proteinfraktionen in Form von Albuminen, Globulinen und Glutelinen gewinnbar (lo), worauf im Folgenden &er eingegangen wird. Material und Methoden Aus Saatgut von Brassica n a p mit Standard-, Einfach-und Doppelqualitat (Vai-ietgten Sollux, Marinus und "DQ"-Neuziichtung) werden im Laboratoriums-mBstab durch Zerkleinerung und Pettextraktion Mehle hergestellt und weiter aufgearbeitet (Schema). Die sukzessiven Extraktionen (Wasser und 5Xige Kochsalzlosungen) werden bei einem Feststoff-Fliissigkeits-VerMltnis von 1:lO durchgefiihrt; die Abtrennung der Feststoffe erfolgt mittels Zentrifugation bei 2500 x g. Der Riickstand der Sdleextraktion wird m c h Waschung mit Wasser bei einem Feststoff-Fliissigkeits-VerhMtnis von 1 : l O und nachfolgender Zentrifugation gefriergetrocknet (Glutelinangereicherte Fraktion) ; ebenso wird mit dern nach isoelektrischer Fallung (pH-Wert 3,5) aus dem Salzextrakt anfallenden gewaschenem Niederschlag (Globulin-Fraktion) verfahren. Der Wasserextrakt w i r d mittels Ultrafiltration in gleiche Anteile Retenat und Filtrat (keine Proteinfallung rnit TrichloressigsWe) fraktioniert . Das Retenat wird spriihgetrocknet bzw. der Diafiltration unterworfen. Dabei wird das anfallende Filtrat periodisch durch Wasser ersetzt, bis ein VerhMtnis von eingesetztem Retenat zu zugegebenem Wasser von 1:2 resultiert. Aus dem UF-Retenat und DF-Retenat werden durch Spriihtrocknung Proteine gewonnen (Albumin-Fraktion). Zur Ultra-und Diafiltration wird eine Ultraf3ltrationsanlage bestehend aus 2 Modulen und einer F'ilterflLiche von 0,08 rn unter Verwendung von Celluloseacetatmembranen Typ CA-M-UF 120 (VEB Zellstoff-und Zellulosewerke Wittenberge) verwendet (11). Die Bestimung des Rohproteingehaltes in den Rohstoffen und Fraktionen erfolgt nach der Halb-Mikro-Kjeldahl-Methode; Isothiocyanate (1%) werden gaschromatographisch und Vinyloxazolidone (VOT) spektrophotometrisch erfdt (12).
Polyenfettsauren der n3-Fettsaurefamilie aus aquatischen Rohstoffquellen, speziell Eicosapentaen-und Docosahexaensaure, spielen bei der alimentaren Prophylaxe und Therapie von Krankheiten des Herz-Kreislauf-Systems eine zunehmende Rolle [I]. In diesem Zusammenhang kommt der Gewinnung von Fettsaurekonzentraten rnit einem hohen summarischen Quotienten an ,,essentiellen" n3-zu n6-Polyenfettsauren besondere Bedeutung zu, wobei die Anreicherung rnit verschiedenartigen physiko-chemischen und/oder chemischen Verfahren praktizierbar ist [2].Zielstellung unter diesem Aspekt durchgefiihrter Untersuchungen war die Ermittlung der Effektivtat einer Harnstoffaddukt-Bildung hinsichtlich einer selektiven Fraktionierung der Fettsauremethylester aus Lipiden von Silberkarpfen (Hypothalmichthys molitrix (Val)), die sich als Seastonfresser gegeniiber anderen limnischen Fischspezies durch einen relativ hohen Anteil an n3-Polyenfettsauren auszeichnen [3]. Material und MethodenDie Gewinnung des Roholes erfolgte aus der Eingeweidepartie von Silberkarpfen mit einer durchschnittlichen Lebendmasse von ca. 2,5 kg durch Kombination von proteolytischem und thermischem AufschluD sowie separative Fettabscheidung [4]. Aus den Roholen wurden die Fettsauremethylester durch basische Umesterung hergestellt [5]. Die Harnstoffaddukt-Bildung wurde in methanolischer Losung vorgenommen; dazu wurde in der ersten Fraktionierungsstufe ein Masseverhaltnis von Fettsauremethylestern zu Harnstoff von 5: 3,4 vorgegeben, und in den nachfolgenden 4 Fraktionierungsstufen wurden jeweils 3,8 g Harnstoff, bezogen auf die nach Abtrennung der Addukte verbleibende flussige Phase, zugesetzt [6]. Die Bestimmung des Fettsiurespektrums in den abgetrennten Harnstoffaddukten erfolgte nach Ausschiitteln mit Diethylether, Trocknen uber Natriumsulfat und diinnschichtchromatographischer Vortrennung an Kieselgel G mittels Hexan/Diethylether/Eisessig = 73/25/2 (v, v, v) als Elutionsmittel durch Gaschromatographie an 15 "/, DEGS auf Chromosorb [7]. Ergebnisse und Diskussion Die Fettsaurespektren der Methylester der eingesetzten Rohole, ihrer Harnstoffaddukte und des nach Adduktbildung resultierenden Finalproduktes finden sich in Tab. 1. o b e r die Veranderungen des summarischen Verteilungsverhaltnisses spezifischer Fettsaurefraktionen rnit unterschiedlichem Sattigungsgrad informieren Abb. 1 und 2. -Durch 5malige Harnstoffaddukt-Fraktionierung der Fettsauremethylester laBt sich erwartungsgemali ein Polyenfettsaurekonzentrat gewinnen, in dem der Anteil an Eicosapentaen-und Docosahexaensaure von 6,5 bzw. 3,4'% auf 23,3 bzw. 27,5% angereichert ist. Damit verbunden ist im weiteren eine drastische Erhohung des Anteils an 18:4 Vertretern der n3-und n6-Fettsaurereihe von 2,3 auf 20,O bzw. 4,O auf 8,5%.Die vorgestellten Untersuchungsergebnisse sind wie folgt interpretierbar :
