Escherichia coli is a frequent cause of lifethreatening bloodstream infections and other common infections, such as urinary tract infections. Antibioticresistant E. coli are readily acquired via the diet (food and water), and other factors such as continuous drug usage, the in appropriate dose usage, and the natural factors of that microbe itself. The resistance results in increased mortality, morbidity, and health care cost. In Indonesia resistant E. coli have emerged, whereas the rates of resistance to some antibiotics were significantly higher, especially to beta-lactam classes (Lestari et al. 2008). Beta-lactams remain the most widely utilized antibiotics owing to their comparatively high effectiveness, low cost, and minimal side effects. Among the various classes of antibiotics, penicillin, and cephalosporins are the most frequently used agents in treatment of bacterial infection. The most common mechanism of resistance to this class of antibiotics is the ability of bacteria to produce beta-lactamases, enzymes that degrade or modify the antibiotic before it can reach the appropriate target site. Therefore, the discovery of a new antimicrobial drug is very important, and the group of rare actinomycetes are really promising as the producer of new bioactive compounds, in this case antibiotics. In this study we screened and characterized the actinomycetes with antibacterial activity against Escherichia coli ATCC 35218 resistant beta-lactam antibiotics. A total of 96 strains collected in Biotechnology Microbial Culture Collection (BioMCC), BPPT, were screened for their antibacterial activities by the agar plug method. Three strains, at-HH-64, at-HH-78, and at-HH-259, showed antibacterial activity. The selected strains were cultured on four different media, both solid and liquid media, e.g. ISP2, ISP4, Micromonospora Starch Medium (MS), and Bennet's Medium (BM), and we characterized their morphology and growth patterns. Morphological characterization showed that all strains belonged to the genera Micromonospora. The active strains were also identified based on 16S rRNA partial sequence. Materi buku diperoleh dari para kontributor seluruh anggota quadhelix dan para narasumber pegiat kecerdasan artifisial di Indonesia. Dalam rangka mengetahui potensi keragaman mikroba Indonesia sebagai sumber senyawa alami, Balai Bioteknologi-BRIN mengoleksikan mikroba dari berbagai lokasi di Indonesia. Selanjutnya, BRIN bekerja sama dengan mitra riset dari Indonesia dan Jepang dalam proyek Science and Technology Research Partnership for Sustainable Development Program (SATREPS) dalam fase I dari tahun 2015 sampai 2020 dan dalam fase II dari tahun 2021 hingga 2026 untuk penemuan senyawa obat dari mikroba. Alur pencarian senyawa obat dari mikroba dimulai dari kegiatan eksplorasi mikroba (isolasi dan identifikasi mikroba) dari berbagai jenis sampel alami (tanah, tumbuhan, serasah, hewan laut, dan lain-lain) melalui waktu yang cukup panjang, yakni antara 10–20 tahun. Salah satu tahap kritis dalam proses penemuan senyawa obat (drug discovery) dari mikroba adalah tahap identifikasi mikroba. Secara umum, proses identifikasi mikroba dilakukan secara molekuler. Namun, proses ini membutuhkan waktu dan biaya yang besar, terlebih bila menangani mikroba dalam jumlah yang banyak dan khususnya mikroba dari golongan fungi yang memiliki bentuk morfologi mikroba tertentu. Proses identifikasi berbasis morfologi dilakukan melalui pengamatan dengan mikroskop. Namun, identifikasi mikroba ini sangat tergantung dari pengalaman yang dimiliki personel pelaksana dan akan menyulitkan apabila mengidentifikasi mikroba yang baru. Kecerdasan artifisial (KA) dapat membantu mengidentifikasi mikroba berbasis morfologi. Dalam hal ini, KA membantu personel dalam menganalisis gambar morfologi suatu mikroba, sehingga proses ini dapat dilakukan secara cepat dan akurat. TKA juga diharapkan dapat membantu personel dalam menemukan mikroba baru yang belum pernah ditemukan sebelumnya, sehingga dapat meningkatkan nilai dari mikroba asli Indonesia tersebut.
