During our search for new antiprotozoal (antimalarial and antitrypanosomal) agents, we have tested culture extracts of soil microorganisms from a variety of sources, including those isolated by the Kitasato Institute for Life Sciences and the Bioresource Laboratories, MicroBiopharm Japan Corporation. We have previously discovered a variety of microbial metabolites exhibiting antiprotozoal, 1,2 antimalarial 3,4 and antitrypanosomal properties. 5,6 Recently, we have isolated a new derivative of pactamycin, designated as jogyamycin (Figure 1), from a culture broth of Streptomyces sp. a-WM-JG-16.2. This compound has proved to have potent antiprotozoal activity (Table 2). Here, we report the fermentation, isolation, structure elucidation and biological activity of this novel aminocyclopentitol antibiotic.The producing organism, strain a-WM-JG-16.2 was isolated from a soil sample collected in Jogjakarta, Indonesia, using the sucrose density gradient centrifugation method. 7 Using the basic local alignment search tool (BLAST), we compared the 16S rRNA gene sequences available in the EzTaxon server to identify the species of strain a-WM-JG-16.2. Streptomyces sp. a-WM-JG-16.2 was classified as a species of the genus Streptomyces; the 16S rRNA gene sequence comparison showing a 100% similarity with S. griseoruber NBRC12873 (AB184209).The strain was cultured on a rotary shaker (220 r.p.m.) at 28 1C for 5 days in 500-ml Erlenmeyer flasks (20 flasks) containing 50 ml of a production medium containing 2% rice starch (Rose brand, Cikampek, Indonesia), 2% glucose (Univar, Sydney, NSW, Australia), 2% soybean meal (J-Oil Mills, Tokyo, Japan), 0.5% yeast extract
Growth Ability of Jatropha Curcas L. Explants on the In Vitro Media Containing IBA and BAProbiotic product is one of the applications of biotechnology that utilize lactic acid bacteria, especially lactobacilli. Some important requirements for microbes that can be used as probiotic include resistance to low pH, ability to grow on bile salts and colonize, and having antimicrobial activity. Each species of the genus Lactobacillus has different characteristics. These characteristics are strongly influenced by the environment in which the bacteria live. This study was carried out in order to characterize Lactobacillus casei which was isolated from dadih. The result of the experiment showed that the isolated L. casei was able to grow on the bile salt at the concentration of 15%, resistant to acid media until pH 2, had antimicrobial activity (significantly inhibited the growth of Escherichia coli, Staphylococcus aureus, and Enterococcus faecalis). Beberapa syarat utama mikroba yang dapat difungsikan sebagai mikroba probiotik antara lain tahan terhadap pH rendah, mampu tumbuh pada garam empedu, mampu berkoloni, memiliki aktivitas antimikroba. Masing-masing spesies dari Genus Lactobacillus memiliki karakteristik yang berbeda-beda. Karakteristik ini sangat dipengaruhi oleh lingkungan dimana bakteri tersebut hidup. Pada penelitian ini telah dilakukan karakterisasi Lactobacillus casei yang merupakan hasil isolasi dari susu kerbau fermentasi. Dari hasil percobaan menunjukkan bahwa L.casei hasil isolasi mampu hidup sampai dengan konsentrasi garam empedu 15%, tahan terhadap media asam sampai dengan pH 2, memiliki aktivitas antimikroba (positif menghambat Escherichia coli, Staphylococcus aureus, dan Enterococcus faecalis). Phenol treatment was applied for isolation of rare Actinomycetes using 25 soil samples collected from Pulau Seribu, Tanjung Redep, Manokwari, and Halmahera. The samples were air-dried and suspended in 1.5 % (w/v) phenol solution at 30 oC for 30 minutes, and subsequently cultured on plates of humic acid-vitamin agar (HVA) medium supplemented with cycloheximide (50 μg/mL) and nystatin (50 μg/mL). A total of 61 isolates were obtained and the most dominant isolates were not Streptomyces (only 24.6%), whereas other genera such as Micromonospora, Actinomadura, Microbispora and Polymorphospora were isolated with ratios of 49.2%, 13.1%, 9.8%, and 3.3%, respectively
Dadih is one of the traditional fermented milk products of West Sumatera. Dadih contain a lot of lactic acid bacteria that acts as a coagulant and preservative. Some lactic acid bacteria also act as a probiotic agent because of characteristics that are resistant to acidic conditions. Some of the main requirements of microbes that can be used as probiotic microbes were resistant to low pH, bile salts, able to colonize, and having antimicrobial activity. Each species of the genus Lactobacillus havedifferent characteristics. This characteristic are influenced by the environment in which the bacteria live. Isolation, identification and characterization of lactic acid bacteria derived from buffalo milk were done. The results of isolated lactic acid bacteria was Lactobacillus plantarum. The characterization of Lactobacillus plantarum indicated that these isolates were able to survive in a concentration of medium containing 0.5% bile salts, resistant to acidic media until pH 2, have antimicrobial activity (inhibit Escherichia coli, Staphylococcus aureus and Enterococcus faecalis).
