OBJETIVO: Delinear os parâmetros que norteiam a escolha de um modelo animal de doença e verificar na literatura biomédica recente quais as espécies animais de uso mais freqüente. MÉTODOS: Considerando a revisão da literatura são discutidos, os conceitos e as características de um modelo animal de doença. Destaca-se o consenso atual sobre quando usar um modelo animal, quais os critérios de sua escolha e os requisitos para um modelo adequado. Descreve quais os tipos de modelos animal de doença e discute as controvérsias da similaridade filogenética e os riscos inerentes a extrapolação dos modelos para os seres humanos. Baseado em uma pesquisa documental na base de dados da BIREME (Medline, Lilacs, Scielo e Biblioteca Cochrane) investiga quais os animais de experimentação mais citados nos artigos destas bases de dados. RESULTADOS: Verificou-se que o rato e o camundongo são os animais mais freqüentemente utilizados. O coelho, cão e o suíno seguem a lista nas referências de língua inglesa. Nas bases de dados da literatura Latino-americana o cão supera o número de citações de coelhos e suínos. Os primatas são minoria nas citações em todas as bases de dados. As revisões sistemáticas também têm no rato o maior número de citações, as demais espécies são citadas em igualdade de condições. CONCLUSÕES: O animal de experimentação é usado virtualmente em todos os campos da pesquisa biológica nos dias de hoje. A relação entre os humanos e os animais de outras espécies ganhou, através dos tempos, contornos mais definidos, a exploração de outras espécies tem regras e uma ética estabelecida, a indução dos resultados do animal para a espécie humana tem critérios claros e objetivos a serem preenchidos. O uso dos modelos experimentais, ou dos modelos animal de doença, ou dos animais de laboratório na pesquisa biomédica permite e deverão permitir nos próximos anos discussões epistemológicas, políticas, sociais, econômicas e religiosas.
OBJETIVO: Verificar, em uma amostra de escolares de uma região de baixo nível socioeconômico, a prevalência de sobrepeso e obesidade. MÉTODOS: Estudo transversal com uma amostra de 218 crianças selecionadas de modo aleatório, a partir de um universo estimado de 1.500 crianças e adolescentes, com idade entre seis e 14 anos, provenientes de três escolas de ensino fundamental de Parelheiros, São Paulo, sem distinção de sexo ou etnia. Foram excluídas aquelas portadoras de qualquer doença metabólica ou endócrina diagnosticada ou em tratamento. Os indivíduos foram submetidos à avaliação antropométrica, com medida do peso corporal e altura para cálculo do índice de massa corpórea e relação peso/altura. Todas as crianças responderam a um questionário aplicado pelos pesquisadores sobre hábitos alimentares. RESULTADOS: A prevalência de obesos e de portadores de sobrepeso foi respectivamente 14,7 e 16,5%. Entre os alunos obesos e com sobrepeso, o baixo consumo de frutas (10,8 e 10,8%), de verduras e legumes (16,4 e 9,1%) e o alto consumo de doces (72,2 e 78,1%) foram associados ao excesso de peso. A prática esportiva esteve ausente ou escassa nos obesos (81,3%) e portadores de sobrepeso (77,8%). CONCLUSÕES: Os alunos avaliados, embora provenientes de uma região pobre de São Paulo, apresentaram um perfil de transição nutricional, com altas taxas de obesidade e sobrepeso.
PURPOSE: To evaluate fecal continence, anorectal manometry (AM) and profilometry (P), in patients operated for congenital megacolon, using either the modified Duhamel technique (MDT) or the modified transanal rectosigmoidectomy (MTR) technique. METHODS: 42 patients were evaluated clinically and via AM and P, for postoperative control. The resting, coughing, voluntary contraction, maintained voluntary contraction and perianal stimulation pressures were investigated. The rectosphincteric reflex was tested and the simple and enhanced pressure curves were evaluated. The three-dimensional profilometric outline was produced. Student's t, chi-squared and Fisher's exact tests were used for statistical analysis (p<0.05). RESULTS: AM showed mean resting pressures of 53.44 mmHg for MDT and 60.67 mmHg for MTR, and mean voluntary contraction pressures of 94.50 mmHg for MDT and 95.47 mmHg for MTR. There was no statistical difference between the groups. The shapes of the simple and enhanced pressure curves did not present any statistical difference, independent of the surgical technique used. CONCLUSION: The two surgical techniques were equivalent. MDT caused greater incidence of postoperative constipation that MTR did. AM and P were shown to be excellent tests for postoperative follow-up among these patients.
OBJETIVO: estudar comparativamente o uso de cianoacrilato e cola de fibrina no reparo do ferimento hepático, para evidenciar suas respectivas capacidades de integração aos tecidos à curto prazo. MÉTODOS: 30 ratos Wistar-EPM1 foram separados em 2 grupos e submetidos à anestesia com tiopental na concentração de 40mg/kg EV. Realizada laparotomia com exposição do fígado. Usando um "punch" dermatológico de 3mm foi produzido um ferimento padronizado em lobo direito hepático. Os animais do grupo A foram tratados com a colocação de cola de fibrina nos ferimentos, os animais do grupo B tiveram os ferimentos tratados com adesivo de cianoacrilato. No 7º de PO, sob anestesia, os animais sofreram a ressecção do lobo hepático direito e foram coletadas amostras para os procedimentos de estudo histológicos (HE e Picro-Sirius). Foram avaliados, por microscopia óptica, os processos inflamatórios (critérios qualitativos da presença de neutrófilos, células gigantes, granuloma tipo corpo estranho, neovascularização) e, por microscopia de polarização, a quantificação de colágeno (Tipo I-jovem e Tipo III-maduro). RESULTADOS: Os dois adesivos levaram ao efeito hemostático em tempos semelhantes. Os ferimentos tratados com cola de fibrina mostraram maior quantidade de colágeno jovem e maduro e uma maior quantidade de neovascularização, enquanto os ferimentos tratados com cianoacrilato mostraram maior reação de granuloma tipo corpo estranho. CONCLUSÃO: O ferimento tratado com a cola de fibrina apresentou maior neovascularização e maior porcentagem de colágeno tipo I e tipo III demonstrando tendência a uma melhor reparação tecidual, sem formação de granuloma tipo corpo estranho.
Occult ischemia of the mobilized stomach is usually related to the dehiscence of an esophagogastric anastomosis. The principle of ischemic conditioning was studied to verify its clinic use. This study aims to evaluate progressively the tissue perfusion of the stomach in ischemic conditioning, establishing the best moment for gastric transposition. Twenty-four male EPM-1 Wistar rats were used, which also underwent partial desvacularization of the stomach by ligature of the left gastric vessels. Tissue perfusion was measured through flowmetry by laser Doppler (tissue perfusion unit; TPU) in the antrum (10 mm distal from the cardiac region). This measurement was done before (baseline) and immediately after the ligature, and on different postoperative days (POD) (days 3, 7, 10, and 14). The study indicates that a larger randomized trial comparing noninvasive positive-pressure ventilation and standard oxygen therapy in children with respiratory failure is feasible, providing a basis for a future trial in this setting. No differences were seen between groups. The number of excluded patients was high.
Capsular contracture is a potential adverse effect of breast implants. An inflammatory reaction is most likely the origin of fibrosis around the implant. It is possible that some substances may act to prevent this inflammatory reaction. Thus, our goal was to evaluate the effectiveness of local depot prednisolone phosphate-liposomes (PPL) on fibrous capsule formation around textured silicone breast implants. Shell prostheses (2 mL) were implanted in the right (plus PPL group) and left (plus saline solution, saline group) subcutaneous dorsum of 18 rats. In another 18 rats, the implants were positioned in the left of the back without any drug instillation (control group). In the PPL group, the capsule thickness (microm) and density (%) of collagen were significantly (p<0.0001) lower compared with the control group on days 35 and 90 postsurgery. Furthermore, in the PPL group, a significant reduction in myofibroblast count was observed on day 90 postsurgery (p<0.0001). In conclusion, a single dose of depot liposome-delivered prednisolone was effective at impairing capsule formation around the silicone implant. The results suggest a strong local and weak systemic effect of PPL on the fibrous tissue around silicone implants. To our knowledge, no study has yet assessed the effect of PPL on silicone breast implants.
