The validation of an analytical procedure for the determination of pesticide residues in beeswax, an interesting matrix for environmental pollution monitoring, is presented. Using the QuEChERS template, the impacts of wax particle size, sample amount, and cleanup procedure (water addition, dispersive solid phase extraction, freeze-out, and combinations thereof) on extraction yield and coextractive load were studied. Sample preparation through liquid-liquid partitioning between acetonitrile and melted wax (∼80 °C), followed by freeze-out and primary-secondary amine dispersive cleanup, was performed on incurred and pesticide-free samples for 51 residues. Determinations were made through LC-MS/MS and GC×GC-TOF, and the whole procedure was validated. Matrix effects were evaluated, with recoveries between 70 and 120% and RSDs below 20% in almost all cases. LC-MS/MS LOQs ranged from 0.01 to 0.1 mg/kg for most pesticides, but for GC-amenable pesticides, GC×GC-TOF sensitivity was lower (0.1-0.2 mg/kg). This methodology can be applied for routine analysis of pesticide residues in beeswax.
Um método analítico empregando cromatografia líquida de alta eficiência com detecção por fluorescência para a determinação simultânea de abamectin e ivermectin em leite bovino foi desenvolvido e validado. Os melhores resultados de recuperação foram obtidos empregando acetonitrila na extração dos compostos, com purificação do extrato por extração em fase sólida em cartuchos contendo C 18 . A derivação pré-coluna foi realizada com N-metilimidazol e anidrido trifluoracético. Os limites de detecção (LOD) do método para abamectin e ivermectin foram 0,10 e 0,14 µg L -1 e os limites de quantificação (LOQ) foram 0,18 e 0,36 µg L -1, respectivamente. As recuperações foram de 75 a 101%, com valores de RSD abaixo de 10%. Os valores de LOD e LOQ obtidos são inferiores aos limites máximos de resíduos em leite estabelecidos pelo Codex Alimentarius, União Européia e legislação brasileira.An analytical method using high performance liquid chromatography with fluorescence detection for the simultaneous determination of abamectin and ivermectin in bovine milk was developed and validated. The best recovery results were achieved by using acetonitrile for extraction of the compounds followed by solid phase extraction in cartridges containing C 18 for the purification of the extract. Pre-column derivatization was accomplished with N-methylimidazole and trifluoroacetic anhydride. The method limit of detection (LOD) values for abamectin and ivermectin were 0.10 and 0.14 µg L -1 and the limit of quantification (LOQ) values were 0.18 and 0.36 µg L -1 , respectively. The recoveries were from 75 to 101%, with RSD values lower than 10%. The LOD and LOQ values are lower than the maximum residue limits (MRLs) in milk established by Codex Alimentarius, European Union and the Brazilian legislation.
A new sensitive, fast and robust method for the determination of paraquat and diquat residues in potatoes, cereals and pulses is presented. Different extraction conditions (solvent, time and temperature) have been evaluated using barley grain, potatoes and dry lentils containing incurred residues of diquat and paraquat. The finalised procedure involves extraction with a mixture of methanol/water/hydrochloric acid at 80 °C and analysis by liquid chromatography-tandem mass spectrometry. Diquat D4 and Paraquat D6 internal standards were added to the test portions prior to extraction. A small-scale inter-laboratory validation of the developed method for diquat and paraquat using potato and barley samples was conducted by three laboratories. The precision and accuracy of the method were determined from recovery experiments (five replicates) at 0.01 and 0.1 mg kg(-1). The recoveries obtained (n = 180) were in the range of 92-120 % with associated relative standard deviation (RSD) between 1.4-10 % for all compound/commodity/spiking concentration combinations.

Assim, a presença de substâncias que são fisiologicamente ativas no metabolismo humano tem sido extensivamente estudada em alimentos. Em vinhos, interesse especial tem sido dedicado às aminas bioativas ou biogênicas e, em particular, à histamina 1 . Embora estas substâncias estejam presentes no vinho geralmente em baixas quantidades, elas exibem interação com o metabolismo humano, fato que justifica a pesquisa. A histamina produz dilatação dos vasos sangüíneos e, conseqüentemente, cefaléia. A eliminação ou a redução desta substância nos vinhos tem sido uma prática cada vez mais utilizada em certos países 2 . A presença de histamina em vinhos está sendo relacionada às más condições higiênico-sanitárias no processo de produção, uma vez que esta substância é formada por algumas espécies de microorganismos. Devido a isso, é importante uma verificação do conteúdo de histamina em vinhos como forma de detectar possíveis falhas no processo de produção 3 . Entretanto, a quantificação de rotina destas substâncias não é utilizada no controle de qualidade dos vinhos, principalmente devido às dificuldades analíticas, como presença de interferentes e complexidade da matriz 4 . As técnicas de separação de aminas empregadas para a determinação de histamina em vinho incluem, principalmente, a cromatografia gasosa 5 (GC, "gas chromatography") e a cromatografia líquida de alta eficiência 6-11 (HPLC, "high performance liquid chromatography"), sendo que as duas técnicas são seletivas e sensíveis, permitindo a detecção de pequenas quantidades de aminas. Neste trabalho desenvolveu-se um método para a determinação de 15 pesticidas em pão utilizando cromatografia gasosa acoplada à espectrometria de massas (GC-MS), operando no modo de ionização química negativa (NCI). No preparo da amostra utilizou-se acetonitrila contendo 1% (v/v) de ácido acético, na presença de acetato de sódio, citrato de sódio e sulfato de magnésio. O método foi validado avaliando os seguintes parâmetros: linearidade, limite de detecção (LOD), limite de quantificação (LOQ), precisão e exatidão. A faixa linear utilizada foi de 1,0-100 µg L -1 . As recuperações foram consideradas satisfatórias apresentando valores entre 70 e 117%, com RSD ≤ 17,6% para quase todos os compostos. O LOQ do método variou de 10 a 20 µg kg -1 . O método demonstrou ser adequado, sensível e altamente seletivo para a análise multirresíduo de pesticidas em pão.In this work a method for the determination of 15 pesticides in bread samples using gas chromatography coupled with mass spectrometry (GC-MS), operating in the negative chemical ionization mode (NCI) was developed and validated. For the extraction procedure acetonitrile containing 1% (v/v) acetic acid was used in presence of sodium acetate, sodium citrate and magnesium sulphate. The method was validated evaluating the following parameters: linearity, limit of detection (LOD), limit of quantification (LOQ), precision and accuracy. The linear range used was from 1.0 to 100 µg L -1 . The recoveries had been considered satisfactory presenting values between 70 and 117% with RSD ≤ 17.6% for almost all the compounds. The real method LOQ ranged from 10 to 20 mg kg -1 . The method shown to be adequate for the multiresidue analysis of pesticides in bread and the GC-MS (NCI) proved to be sensitive and highly selective.
