Peningkatan respon antibodi pascavaksinasi dengan antigen tunggal dan campuran dari bakterin Aeromonas hydrophila and Streptococcus agalactiae diharapkan dapat meningkatkan daya tahan ikan tilapia (Oreochromis niloticus) terhadap penyakit Motile Aeromonas Septicemia/MAS dan Streptococcosis. Sediaan vaksin disiapkan dengan metode pembuatan dan formula yang berbeda, proses inaktifasi dilakukan dengan menambahkan 3% Neutral Buffer Formalin (NBF 10%) pada biakan bakteri dalam media tumbuh BHI dan TSB. Vaksinasi diberikan melalui injeksi intraperitoneal dengan sediaan vaksin monovalen A. hydrophila, monovalen S. agalactiae, dan bivalen A. hydrophila + S. agalactiae (Sel utuh, produk ektraselular/ECP, crude supernatan, campuran sel utuh + ECP, dan broth). Uji tantang dilakukan menggunakan dosis LD50 infeksi tunggal maupun ko-infeksi dari bakteri A. hydrophila dan S. agalactiae. Efektivitas dan keampuhan vaksin tersebut dihitung berdasarkan nilai RPS (Relative Percent Survival) dan hasil respon hematologi. Titer antibodi dapat terdeteksi setelah satu minggu pemeliharaan pasca vaksinasi. Nilai titer antar perlakuan vaksin bivalen berbeda nyata (P<0.05) dengan vaksin monovalen dan kontrol. Nilai RPS vaksin bivalen (campuran sel utuh + ECP) mencapai 100 untuk uji tantang dengan A. hydrophila dan 56,7 pada uji tantang ko-infeksi. Vaksin monovalen A. hydrophila maupun S. agalactiae hanya mampu memproteksi terhadap bakteri homolog, tidak terjadi proteksi silang di antara keduanya.
Penelitian bertujuan untuk mengetahui efektivitas penggunaan vaksin hydrovac dan streptovac untuk pencegahan penyakit bakterial, motile aeromonad septicaemia (MAS) dan streptococcosis pada beberapa jenis ikan budidaya air tawar. Pengujian dilakukan pada skala laboratorium dan lapang. Jenis ikan uji yang digunakan adalah ikan lele, nila, dan gurami. Vaksinasi ikan dilakukan melalui teknik perendaman dengan dosis dan periode sesuai instruksi penggunaan yang tertera pada label produk kedua jenis vaksin tersebut. Efektivitas vaksin dievaluasi berdasarkan pendekatan nilai persen sintasan dan selanjutnya dihitung nilai relative percentage survival (RPS). Hasil penelitian diketahui bahwa nilai RPS vaksin hydrovac pada skala laboratorium pada ikan lele, nila, dan gurami masing-masing sebesar 85,45%; 65,78%; dan 52,28%. Nilai RPS yang dicapai oleh vaksin streptovac terhadap ikan nila sebesar 54,53%. Sementara, nilai RPS vaksin hydrovac pada skala lapang untuk jenis ikan lele, nila, dan gurami masing-masing 70,15%; 52,43%; dan 42,43%; sedangkan nilai RPS yang dicapai oleh vaksin streptovac adalah 40,41%.
Streptococcosis is a significant fish disease impacting tilapia culture in Indonesia, causing losses estimated up to IDR 15.0 billion annually. This study aims to assess the efficacy of bivalent and trivalent vaccines containing Streptococcus agalactiae bacteria on tilapia. The formula of the bivalent vaccine contains 75% of S01-196-16 and 25% of N14G isolates (v/v). Trivalent vaccine contains 30%, 35%, and 35% of N14G, NP1050, and SG01-16 isolates (v/v), respectively. A challenge test assessed the efficacy of the vaccines, and it was carried out at 30, 90, and 150 days post-vaccination by artificially infection at LD60. Selected bacteria isolate to be appointed in the challenge test are N14G (biotype 2) and S01-196-16 (biotype 1). Relative Percentage of Survival (RPS) was used as the main indicator of vaccine efficacy. The results revealed that the highest RPS of a bivalent vaccine against S. agalactiae (S01-196-16) was achieved at the first challenge (61.84%), and trivalent vaccine against S. agalactiae (N14G) and S. agalactiae (S01-196-16) was achieved at the first challenge (61.53% and 76.20%, respectively). Bivalent and trivalent S. agalactiae bacteria vaccines are promising “tools” to control streptococcosis on tilapia.
Toman or Channa micropeltes has high economic potential to be cultured because of their high albumin content. This study aims to obtain information about what kinds of pathogens can infect toman fish in high-density stress (10, 20,30 fish with an average weight of 3 - 15 g and length of 8-14 cm in 20-liter aquarium) and temperatures between 26-34°C that were treated with artificial infections of Aeromonas hydrophila. The results showed that stress treatments with high density and temperature did not cause significant death in toman, or C. micropeltes. The bacteria that can be isolated from toman fish were Aeromonas sp., Moraxella lacunata, Vibrio vulfinicus, and Brevundimonas vesicularis. The parasites found in toman were Trichodina sp. (Prevalence (P) = 25%, Intensity (I) = 1), Gyrodactylus sp. (P= 35%, I= 1.4), Myxosporea sp. (P= 25%, I= 1.2), and Oodinium sp. (P= 20%, I= 7.5), which infected gills, mucus, gut, and skin. Being treated with artificial infection of A. hydrophila, toman fish were very susceptible and 100% death occurred after 24 hours of treatment. The treatment of predisposition factors in toman fish, which can prove adaptation and survival from stress conditions and the presence of the pathogen, will have an impact on the aquaculture development of toman fish so that health management will be applied in aquaculture.
Karakteristik hasil ko-infeksi buatan dari penyakit Streptococcosis dan MAS (Motile Aeromonas Septicemia) dapat dilihat dengan menggunakan parameter gambaran hematologi dan histopatologi. Ikan tilapia (Oreochromis niloticus) ukuran 15 g diinfeksi secara intra peritoneal dengan bakteri Streptococcus agalactiae dan Aeromonas hydrophila menggunakan dosis LD50. Perubahan pertahanan non spesifik ikan terhadap infeksi patogen dilihat dengan mengamati level hematokrit, neutrofil, limfosit, monosit, dan indeks fagositik darah ikan tilapia yang diambil dari arteri caudalis pada hari ke-3, 6, 9, 12, dan 15 setelah infeksi. Hasil analisis perubahan level hematokrit dan limfosit lebih tinggi dibandingkan dengan kontrol, level neutrofil lebih rendah dibandingkan dengan kontrol, dan level monosit dan indeks fagositik fluktuatif selama masa perlakuan memperlihatkan adanya homeostasi gambaran darah ikan terhadap serangan infeksi antigen. Hasil histopatologi organ ginjal, otak, hati, dan limfa memperlihatkan dua pola karakter luka. Pola pertama, luka yang focal sampai terlihat adanya inflamasi dan perdarahan. Pola kedua, luka yang multifocal, luka parah (acute), nekrotik, dan luka inflamasi yang mengakibatkan deformasi sel-sel organ.
