Der postulierte, uber die Zwischenstufen 3, 4 und 5 fiihrende Reaktionsweg wird durch die Isolierung der Zwischenprodukte 4g, 4h und 4i gestiitzt. Diesem zur Pyrimidinbildung fuhrenden Reaktionstyp sind auch bi-und gemischtfunktionelle Reaktionspartner (7, 9) zuganglich.
5-Substituted 4-Aminopyrimidines by Aminomethinylation of AcetonitrilesThe reaction of the s-triazine (1) with the active methylene group of the hetero-and carbocyclically substituted acetonitriles 2a-2i leads to the 5-substituted 4-aminopyrimidines 6a-6i. The postulated reaction path proceeding via the intermediary stages 3, 4, and 5 is supported by the isolation of the intermediates 4g, 4h, and 4i. This reaction type leading to pyrimidine formation comprises also bifunctional reaction components and those carrying mixed functional groupings (7, 9). Die durch den elektrophilen Eingriff des s-Triazins (1) in einen H-aktiven Reaktionspartner initiierte Aminomethinylierungsreaktion fuhrt zu einem Primkprodukt, das als solches isolierbar sein kann, wie im Falle der blausaurefreien Durchfuhrung der Gattermannschen Aldehydsynthesel) oder im System des Pyrazolidin-3,5-dions2). Unter gegebenen Strukturvoraussetzungen kann dieses primk entstandene Aminomethinylierungsprodukt sich jedoch durch Sekundarreaktionen stabilisieren und damit AnlaD zur Bildung von Folgeprodukten geben, beispielsweise bei der Umsetzung von 1 mit Aminothiazolen3), Anilinderivaten4) oder Malonsaureester5).Eine auffallende Sekundarreaktion wurde in der unter Pyrimidinringbildung verlaufenden Umsetzung von 1 mit (3-Pyridy1)-und (2-Fury1)acetonitril gefundens). Die unter der Einwirkung von s-Triazin (1) auf 2,2'-( 1,4-Phenylen)bis-(acetonitril) (2) gebildete aminomethinylierte Zwischenstufe 3 kann in Gegenwart von Morpholin (Sb) durch Iqolierung des Morpholinoacrylonitril-Derivats 6 nachgewiesen werden. Nach zweifacher Aminomethinylierung von 2 erfolgt in Gegenwart sekundarer Amine S Bildungdes Acrylonitriltyps 7. Analoglassensich bei der Umsetzung von 1 mit 5-Amino-4-cyan-1-phenyl-3-pyrazolacetonitril (8) das durch einfache Aminomethinylierung gebildete Intermediarprodukt 9 mittels S in Form der Derivate 11a und l l b , das aus zweifacher Aminomethinylierung hervorgegangene Zwischenprodukt 10 als 12s und 12b, abfangen.
