The aims of the present study were to evaluate the pharmacokinetic profile and efficacy of eprinomectin (EPM) against Rhipicephalus microplus in cattle of a new injectable form of EPM (Voss Performa®). The product was administered subcutaneously at a dose of 200 μg EPM/kg, in a single dose. The efficacy of EPM against R. microplus in cattle was evaluated through field and stall tests. Studies were performed to estimate the pharmacokinetic parameters of EPM with the purpose of better understanding the kinetics of the formulation. The formulation was effective in controlling R. microplus in both naturally and artificially infested cattle, providing efficacy greater than 95%. The results of pharmacokinetic study were Cmax of 47.15 ± 22.20 ng/ml, Tmax of 1.33 ± 0.492 days, T1/2 of 2.96 ± 1.212 days, AUC0–t of 228.08 ± 57.30 ng day ml−1, and AUC0‐∞ of 240.50 ± 58.44 ng day ml−1. Therefore, the new injectable EPM formulation becomes an important alternative for the control of cattle tick in Brazil.
This study was conducted to show the effectiveness of a novel formulation of fipronil in a spot-on formulation for the therapeutic and preventive control of Rhipicephalus sanguineus in naturally infested dogs. Ticks on all dogs were counted at the moment of treatment and weekly after treatment (therapeutic efficacy) or infestation (preventive efficacy). The profile of the therapeutic efficacy for Rhipicephalus sanguineus suggested that the formulation was able to control the Ixodid species for at least 42 days after the treatment.Keywords: Therapeutic and preventive effectiveness, fipronil, Rhipicephalus sanguineus. Foi realizada contagem dos carrapatos no momento da aplicação do produto e semanalmente após o tratamento (eficácia terapêutica), bem como avaliando sua reinfestação (eficácia preventiva). O Complexo das Doenças Respiratórias de Bovinos (CRB) é caracterizado pela infecção do trato respiratório dos animais podendo ser de origem viral, bacteriana ou através da associação de ambos. O CRB tem sido identificado como uma das mais importantes causas de morbidade e mortalidade de criações intensivas, principalmente em animais jovens, causando sérios prejuízos econômicos com o uso de medicamentos para tratamento dos animais e perdas significativas de peso, afetando diretamente a cadeia produtiva da carne bovina. Eprinomectin and praziquantel, nematodicide and cestodicide compounds, are both combined with the insecticide and acaricide compounds fipronil and (S)-methoprene in Frontline® Protect/Broadline®, or esafoxolaner in NexGard® Combo. These topical feline endectoparasiticide products were tested for efficacy against fleas and intestinal helminths in a field trial in Brazil. Flea- and/or helminth-infested domestic cats were treated twice at a monthly interval following label instructions: 160 cats with Frontline® Protect/Broadline® and 165 cats with NexGard® Combo. The flea and intestinal helminth infestations were evaluated using comb counts and copromicroscopy, respectively before first treatment for baseline value, then 9 and 30 days after each treatment for fleas, and 9 days after each treatment for helminths. Multiparasitism was very frequent at baseline, as amongst the 325 included cats, 295, 280, 86 and 93 cats were at least infested with Ctenocephalides fleas, Ancylostoma, Toxocara and Dipylidium caninum, respectively. Efficacies were calculated by comparing the geometric means at baseline and at post-treatment timepoints for each parasite genus/species. Inclusive of both products and of all evaluation timepoints, the Ctenocephalides, Ancylostoma, Toxocara and D. caninum efficacies were at least 98.3%, 99.8%, 99.8% and 96.3%, respectively. No adverse reactions were observed, except for a few instances of mild, transient, and self-resolving hypersalivation occurring on the day of treatment in both groups. This field trial demonstrated high-level efficacy of Frontline® Protect/Broadline® and NexGard® Combo against major parasites of cats in Brazil.
