O objetivo do estudo foi avaliar o efeito antioxidante do extrato hidroetanólico de cipó-mil-homens (Aristolochia tringularis Cham.), em testes in vitro com eritrócitos de indivíduos portadores de doença neurodegenerativa, por meio da avaliação de biomarcadores oxidativos e antioxidantes enzimáticos e não enzimáticos. Realizou-se a análise dos compostos fenólicos totais dos caules de cipó-mil-homens, provenientes da cidade de Panambi- RS. Na sequência foram realizados os testes in vitro, mediante o tratamento com diferentes concentrações do extrato da planta (0,025; 0,050; 0,100 e 0,200 mg/mL). Após, realizou-se a análise dos níveis de TBARS, CAT, GSH e GST. Na dosagem dos compostos fenólicos totais obteve-se o valor de 426mg/mL de extrato. Nas avaliações de estresse oxidativo, foi possível observar o aumento da lipoperoxidação nas concentrações de 0,050mg/mL e 0,200mg/mL, destacando-se a concentração de 0,050mg/mL, onde o dano em lipídeos foi mais expressivo. Não houve alteração significativa nos níveis de GSH em nenhuma das concentrações do extrato utilizadas no estudo. A atividade da enzima catalase mostrou-se diminuída na concentração de 0,100mg/mL. Houve a diminuição da GST nas concentrações de 0,100 e 0,200mg/mL, prejudicando assim a ação antioxidante endógena. Desta forma, o extrato hidroetanólico de cipó-mil-homens não apresentou efeito antioxidante, mas sim um efeito tóxico nessas concentrações.
A esclerose múltipla é uma doença neurodegenerativa, crônica e autoimune que atinge o sistema nervoso central, caracterizada pela desmielinização de neurônios impedindo que os impulsos nervosos cheguem até as áreas pretendidas. O estresse oxidativo trata-se de um desequílibrio entre o sistema de defesa antioxidante e as espécies reativas presentes no organismo. Ambos encontram-se relacionados de alguma maneira que todavia não é elucidada. Visto isto, objetivou-se avaliar o perfil redox de portadores de esclerose múltipla realizando uma comparação com indivíduos saudáveis. Este estudo foi aceito pelo Comitê de Ética em Pesquisa da Universidade de Cruz Alta sob parecer de nº: 3.531.405. Participaram do estudo 10 portadores de esclerose múltipla, grupo (EM) e 8 indivíduos saudáveis, grupo controle (CT) ambos os grupos assinaram o Termo de Consentimento Livre e Esclarecido. Para análises foram coletadas amostras sanguíneas, que foram processadas e foi retirado o plasma. As análises foram realizadas conforme técnicas descritas na literatura. Os resultados foram análisados por teste de Mann-whitney considerando estatisticamente significativo p<0,05. Os resultados encontrados foram: GSH aumentada no grupo EM, Catalase sem diferença estatística entre os grupos, GST aumentada no grupo CT e lipoperoxidação e carbonilação proteíca aumentadas em EM. Conclui-se que os portadores de esclerose múltipla possuem um perfil redox alterado em comparação aos indivíduos saudáveis.

Several in vitro studies have described the erythrocyte toxicity mechanism response to the 2,4-dichlorophenoxyacetic acid (2,4-D) and its metabolites, strongly related to oxidative stress. Compounds such as ascorbic acid and flavonoids, present in various fructiferous plants like Physalis angulata L.- Solanaceae, are exogenous sources of antioxidants, which have aroused interest because of its beneficial biological effects to health by capturing oxygen reactive species. Here, was investigated the cytotoxic effects of the 2,4-D in human erythrocytes exposed to this herbicide and treated with the juice of P. angulata fruits. Analyzing the oxidant and antioxidant mechanisms in these cells in vitro, was demonstrated that the damage mechanism by the Physalis juice occurred at the level of cellular proteins and membranes, altering levels of endogenous antioxidants components such as reduced glutathione and uric acid, and exogenous like vitamin C. Our results indicate new insights into the mechanism of human erythrocytes response exposed to the P. angulata juice, providing wayforward to future studies of cytotoxicity.
