Earlier studies showed that the resistance of cotton genotype FMT 02102996 to Ramularia areola is governed by one dominant gene. More recently, the resistance of another genotype CNPA BA 2003-2059 to R. areola was detected under field and glasshouse conditions. Present investigation was conducted to verify the mechanism of resistance of the genotype CNPA BA 2003-2059 and to find out if the resistance of these two genotypes is governed by the same or by different genes. Segregating plant populations derived from the cross between the resistant genotype CNPA BA 2003-2059 and the susceptible genotype FMT 701, the back cross populations, as well as those derived from the cross between the two resistant genotypes were evaluated for disease severity by artificial inoculations under glasshouse conditions. The ratio of plants segregating for resistance and susceptibility was studied by χ2 test. The results indicated that the resistance to R. areola in genotype CNPA BA 2003-2059 is governed by one dominant gene and that the resistance in each one of the resistant genotypes is governed by a different dominant gene. These results may assist the local breeding programs aimed at pyramiding resistance genes to this pathogen and may form the basis for genetic mapping of resistance genes. RESUMO Mecanismo de resistência e presença de genes diferentes de resistência a Ramularia areola em dois genótipos de algodoeiroA mancha-de-ramulária, causada por Ramularia areola, é uma das doenças de importância econômica para o Brasil. Estudos anteriores demonstraram que a herança da resistência do genótipo FMT 02102996 a R. areola é governada por um gene dominante. Recentemente, a resistência do genótipo CNPA BA 2003-2059 a este patógeno foi verificada em casa de vegetação e no campo. O presente estudo foi realizado para verificar o mecanismo de resistência deste genótipo e verificar se os genes de resistência dos dois genótipos resistentes são os mesmos. Foram avaliadas populações segregantes derivadas do cruzamento entre o genótipo resistente CNPA BA 2003-2059 e o genótipo suscetível FMT 701, dos retrocruzamentos, e também do cruzamento entre os dois genótipos resistentes. As plantas foram classificadas como resistentes ou suscetíveis por meio de inoculação artificial. Com base nos resultados, constatou-se que a resistência do genótipo CNPA BA 2003-2059 é condicionada por um gene dominante e que os dois genótipos possuem um gene diferente de resistência. Estes resultados podem auxiliar o planejamento dos programas de melhoramento do algodoeiro visando piramidar genes de resistência a R. areola em novas cultivares, ao mesmo tempo constituindo informação básica para o início de trabalhos de mapeamento genético deste gene de resistência. A murcha-de-fusário, causada pelo fungo Fusarium oxysporum f.sp. vasinfectum W.C. Snyder & H.N. Hansen, afeta o algodoeiro em qualquer estádio de desenvolvimento. Em plântulas, ocorre amarelecimento, murcha e necrose das folhas cotiledonares; em plantas adultas, ocorre amarelecimento em áreas irregulares da superfície foliar e murcha de folhas e ramos. Algumas plantas afetadas podem sobreviver à doença, emitindo novas brotações próximas ao solo, mas, em geral, os ramos originados a partir desses novos brotos não são produtivos. Durante o processo infeccioso, as plantas perdem todas as folhas e as novas brotações caem. Internamente ao caule, ocorre escurecimento dos feixes vasculares, devido à oxidação e à polimerização de compostos fenólicos. As plantas que conseguem sobreviver sofrem severa redução de crescimento (Davis et al., 2006). O patógeno pode sobreviver no solo na forma de estruturas de resistência (clamidósporos) por períodos superiores a 12 anos (Smith & Snyder, 1975), sendo a RESUMONeste trabalho, descreve-se um método para seleção de genótipos de algodoeiro resistentes à murcha-de-fusário, causada por Fusarium oxysporum f. sp. vasinfectum. Sementes de diferentes genótipos de algodoeiro foram plantadas em bandejas com células (volume de 25 mL), contendo vermiculita esterilizada. Aos 5 e 7 dias após o plantio, depositaram-se, em cada célula, 2 mL de uma suspensão de 5 x 10 5 esporos/mL do patógeno. Avaliou-se diariamente, por 15 dias consecutivos a partir do décimo dia após a inoculação (DAI), a severidade da doença em cada plântula. A resistência dos genótipos foi estimada por meio da área abaixo da curva de progresso da doença (AACPD), além das variáveis calculadas aos 25 DAI: severidade da doença, porcentagem de plântulas com escurecimento do sistema vascular e porcentagem de plântulas com o patógeno interno aos vasos (re-isolamento). Considerando a AACPD, os genótipos Bayou SM-1 e Coker 312 foram mais resistentes que Deltapine 45A. Para a variável severidade aos 25 DAI, além de Bayou SM-1 e Coker 312, o genótipo Auburn 56-24 também foi mais resistente que Deltapine 45A. O genótipo Coker 312 foi mais resistente do que Acala 44 apenas para esta variável. Houve correlação significativa entre AACPD e severidade aos 25 DAI, sendo esta última mais adequada na seleção para resistência, devido à sua praticidade. ABSTRACT. The initial goal of this study was to measure the efficiency of carbon isotope discrimination (Δ) in distinguishing between cotton plant genotypes subjected to two water regimes. In addition, ∆ measurements, leaf water potential and gas exchange ratios were monitored. Using Brazilian breeding lines, this study also tested the usability of ∆ as a proxy for selecting high-performing yield components in cotton plants grown in unfavorable conditions, particularly water deficiency. For these experiments, ∆ and yield components were measured and their correlations analyzed. Differences among cotton genotypes for Δ (p < 0.0001) were verified, and it was found that this variable was significantly correlated with gas exchange. There was a significant positive correlation between Δ and seed cotton yield only in the site experiencing severe water deficiency (Santa Helena de Goiás). However, Δ had a significant negative correlation with fiber percentage. Our results indicate that Δ is a suitable tool for cotton phenotyping, and it may be applied in cotton breeding programs that aim to produce high-performing yield components in unfavorable conditions.
ABSTRACT. With the changes in spinning technology, technological cotton traits, such as fiber length, fiber uniformity, fiber strength, fineness, fiber maturity, percentage of fibers, and short fiber index, are of great importance for selecting cotton genotypes. However, for accurate discrimination of genotypes, it is important that these traits are evaluated with the best possible accuracy. The aim of this study was to determine the number of measurements (repetitions) needed to accurately assess technological traits of cotton genotypes. Seven experiments were conducted in four Brazilian States (Ceará, Rio Grande do Norte, Goiás, and Mato Grosso do Sul). We used nine brown and two white colored fiber lines in a randomized block design with four replications. After verifying the assumptions of residual normality and homogeneity of variances, analysis of variance was performed to estimate the repeatability coefficient and calculating the number of repetitions. Trials with four replications were found to be sufficient to identify superior cotton genotypes for all measured traits except short fiber index with a selective accuracy >90% and at least 81% accuracy in predicting their actual value. These results allow more accurate and reliable results in future researches with evaluating technological traits in cotton genotypes.
