The present study was aimed at investigating the effect of experimental infection by Trypanosoma vivax in different stages of pregnancy, determining the pathogenesis of reproductive failure, and confirming transplacental transmission. We used 12 pregnant ewes distributed into four experimental groups: G1, was formed by three ewes infected with T. vivax in the first third of pregnancy (30 days); G2 comprised three infected ewes in the final third of pregnancy (100 days); G3 and G4 were composed of three non-infected ewes with the same gestational period, respectively. Each ewe of G1 and G2 was inoculated with 1.25 × 105 tripomastigotes. Clinical examination, determination of parasitemia, serum biochemistry (albumin, total protein, glucose, cholesterol, and urea), packed cell volume (PCV), serum progesterone, and pathological examination were performed. Placenta, amniotic fluid, blood and tissues from the fetuses and stillbirths were submitted to PCR. Two ewes of G1 (Ewe 1 and 3) presented severe infection and died in the 34th and 35th days post-infection (dpi), respectively; but both fetuses were recovered during necropsy. In G2, Ewe 5 aborted two fetuses on the 130th day (30 dpi) of pregnancy; and Ewe 6 aborted one fetus in the 140th day (40 dpi) of gestation. Ewes 2 and 4 delivered two weak lambs that died five days after birth. Factors possibly involved with the reproductive failure included high parasitemia, fever, low PCV, body score, serum glucose, total protein, cholesterol, and progesterone. Hepatitis, pericarditis, and encephalitis were observed in the aborted fetuses. The presence of T. vivax DNA in the placenta, amniotic fluid, blood, and tissues from the fetuses confirms the transplacental transmission of the parasite. Histological lesion in the fetuses and placenta also suggest the involvement of the parasite in the etiopathogenesis of reproductive failure in ewes.
Clinical, epidemiological, and pathological aspects of trypanosomiasis caused by Trypanosoma vivax in calves were reported for the first time in northeast Brazil. Clinical and epidemiological data, packed cell volumes (PCV), and parasitemia were assessed in 150 calves in May 2009 (rainy season-survey 1) and in 153 calves in November 2009 (dry season-survey 2) in three farms (A, B, and C). Prevalence of T. vivax in calves examined in the survey 1 was 63.3%, 65.0%, and 80.0% in farms A, B, and C, respectively. Morbidity varied from 63.3% to 80%, mortality from 15% to 30% and lethality from 23% to 37.5%. In survey 1, for all farms, high parasitemia (from 30.3 to 26.2 × 10(6) parasites/mL), fever (from 39.8 to 40.3°C), low PCV (from 15.7% to 18.1%), and body score (from 2.5 to 3.5) were detected. Calves showed depression, weight loss, pale mucous membranes, enlarged lymph nodes, edema of the dewlap, cough, coryza, and diarrhea. The animals from farms A and B were treated with diminazene aceturate. Six months after, in survey 2, non-treated calves from farm C showed values for prevalence (81.82), morbidity (81.82), mortality (12.73), and lethality (15.55) similar to those in survey 1 (P > 0.05). Also in survey 2, four calves aging merely 1-3 days old presented high parasitemia levels (from 32 × 10(6) to 74 × 10(6) parasites/mL), suggesting transplacental transmission. In conclusion, trypanosomiasis by T. vivax constitutes high prevalent disease for calves raised in Brazilian semiarid and may have transplacental transmission.
O maior número de casos de compactação do abomaso ocorreu em bovinos da raça Holandesa com seis casos (42,9%), seguido por animais mestiços com cinco casos (35,8%) e as raças Pardo-Suiça, Nelore e Marchigiana com um caso cada (21,3% Despite the frequent use of dry and fibrous roughage for feeding cows in many regions, especially during the dry season, impaction of the abomasum has been poorly reported in Brazil, probably because the condition is misdiagnosed by practitioners. The present paper aimed to accomplish a retrospective study on primary abomasal impaction in 14 cattle from Pernambuco State, northeastern Brazil. Eight moderate cases, without severe abdominal distention and with no rumen compaction, were treated conservatively, and four severe cases, with severe abdominal distention and rumen compaction, were treated surgically. One bull was slaughtered and one cow died without treatment. The greater number of abomasal impaction cases was in Holstein cows with six cases (42.9%), followed by crossbred cattle with five cases (35.8%), and the breeds Brown-Swiss, Nelore and Marchigiana, each with one case (21.3%). The food composition was characterized by low quality fibers and varied greatly among cases. Most frequent clinical signs were apathy, dehydration, hypomotility and ruminal bloat, intestinal hypomotility and scanty or absent feces with mucus. The hematological findings revealed leukocytosis with neutrophilia and hyperfibrinogenemia in most cases. Ruminal fluid analysis showed compromised flora and fauna dynamics and increased chlorine ion concentration. Clinical (4/8) and chirurgical (2/4) recovery rate achieved 50%. The clinical and chirurgical methods remain as viable options for the treatment of light and severe impaction, but the prognosis is always reserved especially when associated to late pregnancy.
233Pesq. Vet. Bras. Three outbreaks of poisoning by Prosopis juliflora pods are reported in the semiarid region of the state of Pernambuco, Northeastern Brazil, in cattle grazing in fields invaded by the plant or ingesting mesquite beans as a concentrate food. In two farms the disease occurred sporadically. In another, 112 (9.28%) cattle out of 1206 were affected, 84 (6.96%) died due to emaciation, and 28 (2.32%) gained weight after the pods had been withdrawn from the feed. Main clinical signs were progressive weight loss, atrophy of the masseter muscles, dropped jaw, tongue protrusion, difficulties in prehending food, tilting the head during mastigation or rumination, salivation, impaired swallowing, and decreased tone of the tongue. The hematology reveals hypoproteinemia and anemia. Gross lesions were emaciation and reduction in size of the masseter muscles, which appear thinner than normal and grayish due muscular atrophy. Degeneration of neurons of the trigeminal motor nuclei, Wallerian degeneration of the trigeminal nerve roots, and muscular atrophy of the masseter muscles with substitution by fibrous tissue were observed on histologic examination. For the prevention of the poisoning is necessary to limit the amount of mesquite beans in animal nutrition. It is also necessary to develop research to determine the economic and sustainability of the use of Prosopis juliflora for animal food, human food or other uses such as charcoal, wood and fuel wood. This study aimed to describe the toxic effects resulting from the administration of Calotropis procera (Aiton) W. T. Aiton latex to rats and C. procera leaves to sheep. We studied male sheep that received C. procera leaves by gavage. Twenty male rats were separated into 5 groups and were subjected to an intra-peritoneal injection of fresh C. procera latex (without carrier solvent) at 1.0, 0.6, 0.3 or 0.1 ml of latex/kg of body weight, and control animals were injected with 0.9% NaCl. All rats were treated with the highest dose, but none of the rats from the other groups, died. The histological lesions were restricted to rats dosed with 1.0 ml of latex/kg body weight and included multi-focal coagulation necrosis of cardiac fibers and vacuolized hepatocytes. Subsequently, three groups of two sheep were treated with (1) a single dose of 30 g/kg, (2) a single dose of 60 g/kg or (3) 60 g/kg per day for 10 consecutive days. Exposure to the C. procera leaves was responsible for tachycardia and transitory cardiac arrhythmias in sheep from all groups. Gross pathological analysis of sheep dosed with 60 g/kg per day for 10 days revealed mild ascites, exudates on the trachea, pulmonary edema, mild hemorrhage in the liver, hydropericardium, flaccid heart, ulcers on the abomasum and kidneys presenting pale juxtamedullary cortex. The histological findings of the rat and sheep studies were similar and included multi-focal coagulation necrosis of cardiac fibers and vacuolized hepatocytes. In conclusion, our findings indicate that C. procera is a cardiotoxic and hepatotoxic plant.
Gossypol, a polyphenol compound produced by cotton plant, has proven reproductive toxicity, but the effects of gossypol on sheep ovaries are unknown. This study was aimed to determine the in vitro and in vivo effects of gossypol on the ovarian follicles of sheep. This trial was divided into two experiments. In the first one, we used twelve non-pregnant, nulliparous, Santa Inês crossbred ewes, which were randomly distributed into two equal groups and fed diets with and without cottonseed cake. Feed was offered at 1.5% of the animal’s body weight for 63 days. The concentrations of total and free gossypol in the cottonseed cake were 3.28 mg/g and 0.11 mg/g, respectively. Throughout the trial period, no animal showed clinical signs of toxicity and no effects on body weight were observed. However, there was a significantly lower number of viable ovarian follicles (20.6%) and higher number of atretic follicles (79.4%) in the gossypol-fed sheep compared to the control (85.1 and 34.9%, respectively). These findings were observed at all stages of follicular development. In the second experiment, eight ovaries from slaughterhouse were cultured with different concentrations of gossypol acetic acid (0, 5, 10 and 20 μg/mL) for 24 hours or seven days. The in vitro action of gossypol resulted in a significant decrease in viable ovarian follicles, especially the primary and transition follicles, and a significant increase in the number of atretic follicles after 24 hours of culture. These follicles were greatly affected when cultured with gossypol for seven days. It is concluded that gossypol present in cotton seeds directly acts on ovarian follicles in sheep to increase atresia.
Pesq. Vet. Bras. The aim was to conduct a retrospective study of the main epidemiological, clinical, pathological and laboratory ϐindings from sheep suffering from obstructive urolithiasis, in the Agreste region of Pernambuco, Brazil. Thus, 66 medical records of non-castrated male sheep with age from 2 months and 5 years were analyzed. Most cases (79.63%) occurred during the rainy season. Most sheep were raised intensively (87.88%) and all were fed with concentrated ration. Main clinical signs were manifestation of pain, congestion of the mucosae, hyperthermia, and increased abdominal tension associated with urinary tract obstruction. Half of the animals showed total obstruction of the urethra, 69.7% of which died, whilst in cases of partial obstruction only 30.3% died. Laboratorial exams revealed neutrophilia and regenerative left turn, hyperϐibrinogenemia, and azotemia. Urinalysis showed hematuria in 89.29% of the cases, and acidic pH in 46.43%. Erythrocytes, leukocytes, cell desquamation of urethral epithelium, and crystals of amorphous urate predominated in the examination of the sediment. Most frequent gross ϐindings were pyelonephritis (61.54% of cases) and hydronephrosis (50%). In the ureter, ureteritis and hydroureter was observed. The bladder showed diffuse hemorrhagic cystitis (50%) and uroliths (57.69%). In the urethra, uroliths (61.54%) and diffuse hemorrhagic urethritis (57.69%) was found. The uroliths were composed mainly by hydrated calcium oxalate. It is concluded that in the Agreste region of Pernambuco, urolithiasis is an important disease of sheep with high mortality, associated with the ingestion of large amounts of concentrates and mineral supplements.
925Pesq. Vet. Bras.
