RESUMO -O envolvimento do sistema nervoso central na paracoccidioidomicose vem ganhando importância dentro do espectro de manifestações clínicas da doença. O primeiro registro na literatura de que o Paracoccidioides brasiliensis seria também capaz de comprometer órgãos do sistema nervoso central foi descrito em 1919 por Pereira & Jacobs; desde então muitos outros estudos têm demonstrado esta forma de apresentação clínica, sendo que em alguns sua frequência atinge os 27,27%. Descrevemos o caso de uma paciente de 34 anos, residente em Juiz de Fora-MG, admitida com quadro clínico compatível com pneumonia e que durante a internação iniciou sintomatologia cerebelar (náuseas, vômitos, ataxia de marcha e dismetria). A tomografia computadorizada de crânio revelou lesão cerebelar sugestiva de abscesso. O tratamento proposto baseou-se na exerese cirúrgica da lesão e, como a anátomo-patologia mostrasse presença de leveduras em gemulação múltipla ("roda de leme de navio"), diagnóstico de neuroparacoccidioidomicose pôde ser firmado, e o tratamento complementado pela associação de sulfametoxazol-trimetoprim. A paciente evoluiu bem, persistindo discreta paresia e hipoestesia em membro superior direito. PALAVRAS-CHAVE: paracoccidioidomicose, sistema nervoso central, neuroinfecção. Paracoccidioidomycosis of the central nervous system: case reportABSTRACT -The involvement of the central nervous system in paracoccidioidomycosis is more frequent than previously thought. The first reference to the possibility that Paracoccidioides brasiliensis could affect the central nervous system was by Pereira & Jacobs in 1919. Since then, a great number of other studies has showed this form of clinical behavior and, in some of them, the frequency has ranged 27.27%. We report a clinical case of a 34-year-old white Brazilian woman admitted because of bacterial pneumonia. In the sixth day of admission, the patient developed cerebellar symptomatology with nausea, vomiting, dysmetria and gait disturbance. Central nervous system computer tomographic scanning disclosed a hypodense lesion in the right cerebellar hemisphere. The patient was submitted to surgery with total excision of the lesion. Histopathological examination confirmed the diagnosis of neuroparacoccidioidomycosis. Coadjuvant treatment with sulfamethoxazole-trimetoprim was introduced. Resumo: FUNDAMENTOS: O câncer de pele é o mais frequente tipo de câncer humano e mostra aumento de sua incidência. Em muitos casos, antes do surgimento do carcinoma, instala-se uma lesão precursora, ceratose actínica, podendo evoluir para carcinoma espinocelular. Estudos buscam determinar os parâmetros com significado prognóstico na predição daqueles tumores que terão comportamento mais agressivo. OBJETIVO: Avaliar a expressão dos marcadores de proliferação celular (PCNA, Ki-67) e apoptose (p53, Bcl-2), em portadores de carcinoma espinocelular e ceratose actínica. MÉTODO: Foram estudadas amostras de 30 pacientes: sendo dez portadores do carcinoma espinocelular; dez de ceratose actínica e dez indivíduos livres de lesões submetidos à blefaroplastia. RESULTADOS: A proteína p53 foi expressa em todos os casos estudados, embora apresentassem padrões quantitativos diferentes. O Bcl-2 foi expresso em baixa intensidade. Em seis casos de ceratose actínica, nas peles de blefaroplastia, e negativo nos casos de carcinoma espinocelular. O PCNA exibiu expressão intensa, em todas as amostras. O Ki-67 apresentou expressão variável, nos casos de carcinoma e de ceratose, e negativo na pele de pálpebra. CONCLUSÃO: A expressão do Ki-67 e a não-expressão de Bcl-2, no grupo CEC, indica intensificação da atividade proliferativa. Ao passo que, a maior expressão de p53 e Bcl-2, no grupo CA, sugere imortalização celular. Palavras-chave: Apoptose; Carcinoma de células escamosas; Ceratose; Imuno-histoquímica; Proliferação de células Abstract: BACKGROUND: Skin cancer is the most frequent type of human cancer and has shown an increase in its incidence. In many cases, before the onset of the carcinoma, there might be a precursor lesion -actinic keratosis, which can develop into squamous cell carcinoma. Studies have been carried out in order to determine the parameters that have prognostic significance in predicting those tumors which have more aggressive behavior. OBJECTIVE: To evaluate the expression of markers of cell proliferation (PCNA, Ki-67) and apoptosis (p53,Bcl-2) in patients with squamous cell carcinoma and actinic keratosis. METHOD: We studied samples from 30 patients, ten patients of squamous cell carcinoma, ten with actinic keratosis and ten lesion-free samples from blepharoplasty. RESULTS: p53 protein was expressed in all cases with different quantitative patterns. Bcl-2 was expressed at low intensity in six cases of actinic keratosis in the skin from blepharoplasty and negative in cases of squamous cell carcinoma. PCNA showed intense expression in all samples. Ki-67 showed variable expression in cases of keratosis and carcinoma and negative in the skin from the eyelid. CONCLUSION: The high expression of Ki-67 associated with low expression of Bcl-2 indicates proliferation in the carcinoma group. Thus, expression of p53 and Bcl-2 in patients with actinic keratosis indicates cell immortalization.
- Basal cell carcinoma is the most common form of human cancer. Studies employing molecular and genetic biology techniques, associated with histomorphology, lead to the identification of risk factors in the development of more recurring and aggressive lesions. OBJECTIVE - To correlate markers expression of apoptosis (p53 and bcl-2) and cell proliferation (Ki-67 and PCNA) with histological indicators of tumor severity. METHODS - Five samples of the nodular, morpheaform and superficial types of carcinoma were studied, respectively.One control group with three lesion-free patients was also included in the study. The Mann-Whitney test was used to compare these markers expression with the manifestation form of basal cell carcinoma. RESULTS - Bcl-2 expression was significant in basal cell carcinomas said to be aggressive (morpheaform and nodular types). Of the studied tumors, 66.7% (n =10) strongly expressed p53.Our results show a greater expression of Ki-67 in nodular and superficial basal cell carcinoma, with no expression in the controls. PCNA showed a strong expression in all types of tumors and in the controls. CONCLUSION - The findings allow us to conclude that Bcl-2 and p53 show a tendency to indicate the severity of basal cell carcinoma. In contrast, Ki-67, due to its variable behavior, cannot be considered a marker of severity. Also, PCNA was not a good marker of cell proliferation.
The Brazilian Meeting on Research Integrity, Science and Publication Ethics (BRISPE) has been held every two years since 2010 in Brazil. The BRISPE has been the major forum for discussion of research integrity in the country. Although it is a national meeting, it has strengthened the role Brazilian science has played in international conversations on the responsible conduct of research. In 2010, the I BRISPE (www.ibrispe.coppe.ufrj.br) focused on research integrity issues related to research projects, to the submission and review process of manuscripts, and to authorship. In 2012, the II BRISPE (www.iibrispe.coppe.ufrj.br) addressed research integrity and leadership in science. The meeting looked at the country's responsibility to foster responsible research, considering its scientific leadership in Latin America. The meeting led to the publication of the Joint Statement on Research Integrity, (www.iibrispe.coppe.ufrj.br/ images/IIBRISPE/JoinStatement/JointStatementonResearchIntegrity_IIBRIS-PE_2012_English.pdf). In 2014, the III BRISPE (http://www.fapesp.br/8788) focused on institutional policies to foster research integrity initiatives at universities and research centers in Brazil. Locally, the III BRISPE was also a preparatory meeting for the 4 th World Conference on Research Integrity, which was held in Rio de Janeiro, in 2015 (www.wcri2015.org). In 2016, the IV BRISPE (www.brispe2016.org) drew upon the role of mentors, editors and funders to strengthen a research integrity culture in Brazilian science. The event was held at the Federal University of Goiás (UFG), November 17-18, 2016, with 260 participants. For this fourth edition of the meeting, there was a call for submissions for oral and poster sessions on research and education and on science policy. From 40 submissions, 34 were accepted. The Proceedings of the IV BRISPE contain the abstracts of oral and poster presentations of authors who agreed to have them published. The V BRISPE will be held in 2018.

Introdução: Procedimentos cirúrgicos com oclusão da aorta têm sido associados a lesão pulmonar de reperfusão. O objetivo deste trabalho foi estudar a presença de edema pulmonar em um modelo de lesão de reperfusão por oclusão da aorta em ratos. Material e métodos: 33 ratos Wistar foram anestesiados com tiopental sódico (20mg/kg, via intraperitoneal), seguindo-se laparotomia por incisão mediana, para isolamento da aorta e cava inferior. A seguir, os animais foram randomizados em três grupos. Grupo isquemia-reperfusão (GIR, n = 5): animais submetidos a 30min de oclusão dos vasos, seguindo-se 120min de reperfusão. Grupo isquemia (GI, n = 5): animais submetidos a 30min de oclusão dos vasos. Grupo controle (GC, n = 5): animais submetidos apenas ao procedimento cirúrgico, sem oclusão dos vasos, acompanhados por 150min. Após o sacrifício, em 15 animais foram realizados estudos histopatológicos dos pulmões. Para caracterização de edema, foi realizada a análise morfométrica por contagem de pontos, determinando-se o índice de edema alveolar. Realizou-se também uma análise semiquantitativa da infiltração de polimorfonucleares nos pulmões. Em 18 animais retirou-se o pulmão direito para a determinação da relação entre os pesos úmido e seco. O índice de edema alveolar e a relação peso úmido/peso seco foram comparados entre os grupos através de ANOVA, com a correção de Bonferroni para comparação entre os grupos dois a dois. Resultados: Os animais do GIR apresentaram maior edema alveolar em relação aos do GI e GC (0,24; 0,18; 0,17; respectivamente, com p < 0,001). Não houve diferenças nas relações peso úmido/peso seco dos três grupos. Houve maior infiltração de células inflamatórias nos pulmões dos ratos submetidos a isquemia-reperfusão. Os autores concluem que a oclusão e desoclusão da aorta infra-renal está associada à lesão pulmonar. Essa lesão não foi induzida pela elevação da pressão hidrostática na circulação pulmonar em função da oclusão da aorta, pois ela não foi encontrada no grupo submetido apenas à isquemia.
