Abstract:The passion fruit (Passiflora edulis Sims), also known as the purple granadilla, is widely cultivated as the new darling of the fruit market throughout southern China. This exotic and perennial climber is adapted to warm and humid climates, and thus is generally intolerant of cold. There is limited information about gene regulation and signaling pathways related to the cold stress response in this species. In this study, two transcriptome libraries (KEDU_AP vs. GX_AP) were constructed from the aerial parts of cold-tolerant and cold-susceptible varieties of P. edulis, respectively. Overall, 126,284,018 clean reads were obtained, and 86,880 unigenes with a mean size of 1449 bp were assembled. Of these, there were 64,067 (73.74%) unigenes with significant similarity to publicly available plant protein sequences. Expression profiles were generated, and 3045 genes were found to be significantly differentially expressed between the KEDU_AP and GX_AP libraries, including 1075 (35.3%) up-regulated and 1970 (64.7%) down-regulated. These included 36 genes in enriched pathways of plant hormone signal transduction, and 56 genes encoding putative transcription factors. Six genes involved in the ICE1-CBF-COR pathway were induced in the cold-tolerant variety, and their expression levels were further verified using quantitative real-time PCR. This report is the first to identify genes and signaling pathways involved in cold tolerance using high-throughput transcriptome sequencing in P. edulis. These findings may provide useful insights into the molecular mechanisms regulating cold tolerance and genetic breeding in Passiflora spp.

A concise and stereoselective total synthesis of (±)-cephanolide B was achieved in 15 steps. The key steps in the synthesis were as follows: (i) an intermolecular Diels–Alder reaction followed by lactonization to form the oxabicyclo[2.2.2]­octane DE ring; (ii) a tandem reaction, featuring an intramolecular Pauson–Khand reaction, a 6π-electrocyclization, and an oxidative aromatization by O2, to construct the ABC-tricyclic rings (6-5-6); and (iii) a phthaloyl peroxide-mediated arene oxygenation to install the C-13 phenol group.

Background Passiflora edulis , known as passion fruit and native to South America, is now widely cultivated throughout southern China for its edible value, medicinal efficacy and ornamental properties. We have developed a cold-tolerant variety of P. edulis (‘Pingtang 1’) that can survive subzero temperatures and is highly adaptable in Karst areas. In this study, cuttings of ‘Pingtang 1’ were cultivated in a limestone (L) rocky desertification area and a sandy dolomite (D) rock desertification area. Changes in nutrient elements in both the soils and plants were revealed in the two plots. Moreover, RNA sequencing (RNA-Seq) was performed to profile the root transcriptomes for further exploration of nutrient adaptative mechanism of Passiflora edulis in Karst regions. Results In this study, a total of, 244,705,162 clean reads were generated from four cDNA libraries and assembled into 84,198 unigenes, of which 56,962 were annotated by publicly available databases. Transcriptome profiles were generated, and 1314 unigenes (531 upregulated and 801 downregulated) were significantly differentially expressed between the L and D root cDNA libraries (L_R and D_R, respectively); these profiles provide a global overview of the gene expression patterns associated with P. edulis adaptability to Karst soils. Most unigenes including a number of differentially expressed genes (DEGs) were involved in nutrient element uptake, utilization, signal regulation. And DEGs enriched in KEGG pathways of plant hormone signal transduction, phenylpropanoid biosynthesis, and biosynthesis of unsaturated fatty acids were significantly expressed. Conclusion These results could contribute to better understanding the adaptation of this species to environmental stress and thus enhance the potential for successfully introducing and commercially deploying P. edulis . This study assesses spatial and temporal variation in environmental variables in relation to phytoplankton community size and composition in a typically eutrophic river reservoir (Hai River, northern China). The aim is to identify environmental parameters governing spatial and temporal differences in phytoplankton density and composition. Physicochemical parameters, including nutrient concentrations, were determined in monthly surface water samples from 2015. The average concentration of key eutrophication indexes (i.e., total phosphorous (TP: 0.24 ± 0.11 mg·L −1 ), total nitrogen (TN: 2.96 ± 1.60 mg·L −1 ), and Chlorophyll a (Chl a: 38.5 ± 11.5 mg·m −3 )) substantially exceeded threshold values for eutrophic streams. Moreover, the eutrophication increased significantly downstream along the river reservoir as a consequence of an increasing fraction of agricultural and industrial land-use in the watershed. 103 phytoplankton species were identified, of which Chlorophyta was the dominated phylum (47 species), followed by Bacillariophyta (23 species) and Cyanophyta (18 species). No spatial difference in species distribution (ANOVA, p > 0.05) were found, while the temporal differences in species composition exhibited significant heterogeneity (ANOVA, p < 0.001). Phytoplankton abundance was highest in early summer (June and July), with maximum values increasing from 1.78 × 10 8 and 2.80 × 10 8 cells·L −1 in upstream and middle reaches, respectively, to 4.18 × 10 8 cells·L −1 furthest downstream. Cyanophyta, also known as Cyanobacteria and commonly referred to as blue-green algal, are known to constitute algae bloom in eutrophic systems. Common species are Microcystis marginata, Microcystis flos-aquae, and Oscillatoria sp. This was the dominant phyla during summer months, especially in the middle and lower reaches of the stream reservoir where it accounted for 88.9% of the phytoplankton community. Shannon weaver index (H') and Pielous's evenness index (J') were extremely low (1.91-2.43 for H' and 0.39-0.45 for J') in samples collected from the lower part of the stream during the period of algal bloom, indicating an imbalance in the phytoplankton communities. Canonical correspondence analysis (CCA) indicated that water temperature (WT) and possible pH, along with nitrate (NO 3 -N) and nitrite (NO 2 -N), were the most important explanatory parameters in regard to phytoplankton composition. This research provides an understanding of the role of physicochemical water quality parameters in governing algal blooms and phytoplankton composition in river reservoirs.

Photoredox‐catalyzed isomerization of γ‐carbonyl‐substituted allylic alcohols to their corresponding carbonyl compounds was achieved for the first time by C−H bond activation. This catalytic redox‐neutral process resulted in the synthesis of 1,4‐dicarbonyl compounds. Notably, allylic alcohols bearing tetrasubstituted olefins can also be transformed into their corresponding carbonyl compounds. Density functional theory calculations show that the carbonyl group at the γ‐position of allylic alcohols are beneficial to the formation of their corresponding allylic alcohol radicals with high vertical electron affinity, which contributes to the completion of the photoredox catalytic cycle.

The ability of a new SBR (sequencing batch reactor) based on simulating photovoltaic aeration for maintaining nitrification activity under a 25-day starvation period was studied. The activity and abundance of ammonia-oxidizing bacteria (AOB) and nitrite-oxidizing bacteria (NOB) and the diversity of AOB were investigated. The measured biomass decay rates were 0.017 day(-1) and 0.029 day(-1) for AOB and NOB, respectively. These decay rates correlated well with AOB and NOB population quantified by real-time PCR. The recovery of ammonia oxidation rate and nitrite oxidation rate needed 4 and 7 days, respectively, indicating that NOB was more affected than AOB by starvation conditions. According to the real-time PCR results, Nitrospira was the dominant NOB in the reactor. Phylogenetic analysis indicated that Nitrosomonas oligotropha cluster was the dominant major cluster before and after starvation. Moreover, Pareto-Lorenz evenness distribution curves were plotted to interpret the interspecies abundance of AOB; the results suggested that AOB community possessed a balanced structure with medium Fo (Functional organization). Thus, the community can potentially deal with changing environmental conditions (e.g., starvation) and preserve its functionality according to the concept of functional redundancy.
