Two-Step Optimization to Develop a Transdermal Film Loaded With Dapoxetine Nanoparticles: A Promising Technique to Improve Drug Skin Permeation

Abstract: Dapoxetine (DPX) is an orally administered drug for the treatment of premature ejaculation (PE). One of the challenges of administering DPX orally as a tablet is its poor bioavailability (ie, 42%) due to extensive first-pass metabolism. Thus, it is vital to develop a new formulation and mode of delivery to achieve the unmet needs of PE treatment. In this study, an optimized DPX polymeric nanoparticle (PNP) was developed and subsequently loaded into a transdermal film. The Box–Behnken design was utilized to opt… Show more

“…Nanotechnology, through the development of NPs, has been revolutionizing drug delivery, since its encapsulation significantly improves the bioactive release, absorption, and safety profile of loaded compounds or extracts [ 28 , 29 ]. Despite its success, the formulation of NPs containing bioactive compounds from seaweeds is still a largely unexplored topic [ 30 ].…”

“…One string was attached with this snare, which disregarded the pulley and a little dish appended to the opposite end for holding the weight. Little pointer was appended to the string, which goes over the scale attached on the base plate [25]. For estimation of tensile strength, patch was pulled and loads were bit by bit added to the dish to build the pulling power till the patch was broken.…”
