The use of polyvinyl alcohol glutaraldehyde antigen coated discs for laser induced fluorescence detection of plague

Abstract: Discs of polyvinyl alcohol cross-linked with glutaraldehyde were synthesized under acid catalysis (H 2 SO 4 ). Then, the antigen F1 purified from Yersinia pestis was covalently linked to this modified polymer. Afterwards, an enzyme-linked immunosorbent assay (ELISA) was established for the diagnosis of plague in rabbit and human. The best conditions for the method were achieved by using 1.3 µg of F1 prepared in 0.067 M phosphate buffer, pH 7.2, containing 1 M NaCl (PBS); anti-IgG peroxidase conjugate diluted … Show more

“…Due to its biocompatability, PVA could also be used in a variety of biomedical applications. 3,10,11 A PVA gel membrane could be used as a synthetic membrane for pervaporation and as a bioartificial pancreas in biomedical applications. 4 -11 The reaction between PVA and glutaraldehyde (GA) generates a polymer network, whose mechanical properties and swelling are affected by its degree of crosslinking.…”

“…In our laboratories other procedures for antigen covalent immobilization on supports have been proposed such as Dacron (11), ferromagnetic Dacron (12,13), interpenetrated network of PVA-glutaraldehyde (14)(15)(16)(17), filter paper plasticized with PVA-glutaraldehyde (18), and a polyaniline-Dacron composite (19). The optimal values of F1 antigen amount, conjugate dilution and serum titer established for the present method (1.25 µg/disc; 1:4,000 and 3,200, respectively) can be compared to those found for PVA-glutaraldehyde (1.3 µg/disc; 1:6,000 and 1:12,600), plasticized filter paper (1.25 µg/disc; 1:12,000 and 1:6,400), and polyaniline-Dacron (2 µg/disc; 1:4,000 and 1:800).…”
