The role of endothelin-1 and endothelin receptor antagonists in allergic rhinitis inflammation: ovalbumin-induced rat model

Abstract: ET-1 is one of the mediators that impact AR development and ET-1 antagonists can be useful for symptom control and for decreasing allergic inflammation in AR patients.

“…Antihistamines, which have an effect on all major symptoms, have been evaluated in many previous studies and their histopathological effects on the nasal mucosa have been demonstrated. In a previous study using antihistamines by Tatar et al 17 observed that edema decreased, damage to the basal membranes of the respiratory epithelium, continued cilia loss, decreased inflammatory cells, increased fibroblasts in the lamina propria, and formation of collagen fibers. Another study, 18 examined the effect of intranasal azelastine on the nasal mucosa; eosinophil and mast cell infiltration decreased, inflammatory cells decreased and there was focal goblet cell metaplasia on the respiratory epithelial surface.…”

“…Furthermore, the effectiveness of bosentan has been reported in several mouse or rat disease models. For example, in an ovalbumin‐induced allergic rhinitis rat model, oral bosentan significantly inhibited nasal symptoms and serum IgE level 37 . In addition, in a murine model of inflammatory bowel disease and a rat colitis model, bosentan significantly reduced clinical and histological inflammation 38,39 .…”

“…The liver tissues (20 mg) were stabilized in an RNA stabilization reagent (RNAlater, Qiagen) and then disrupted with the TissueLyser II (Qiagen) (2x2 min). All of the RNA was purified using an RNeasy Mini Kit (Qiagen) and RNA samples were reverse-transcribed into complementary DNA with a high-capacity cDNA reverse transcription kit (Qiagen) according to our previous study [16,17] .…”
