RESUMO: "Citotoxidade de extratos e constituintes de Wedelia paludosa D.C." Wedelia paludosa D.C. (Asteraceae) é uma planta ornamental facilmente encontrada em várias regiões do Brasil, principalmente nos estados de Santa Catarina, São Paulo, Minas Gerais, Bahia e Pernambuco. Objetivando descobrir novas substâncias citotóxicas a partir desta espécie, o extrato hidrometanólico de W. paludosa (HME) e as frações diclorometânica (FD) e aquosa (FA) resultantes de sua partição em CH 2 Cl 2 -H 2 O foram avaliados utilizando-se o bioensaio em Artemia salina. A fração diclorometânica (FD) apresentou a maior atividade citotóxica (CL 50 = 140,6 mg/mL), e sua análise por cromatografia líquida de alta eficiência empregando-se fase reversa (FR-CLAE) revelou os ácidos caurenóico (1, 6,22 ± 0,23%) e grandiflorênico (2, 3,22 ± 0,31%) como constituintes majoritários. As amostras HME (CL 50 = 980 mg/mL), FD (CLC 50 = 140,6 mg/mL), 1 (CL 50 = 15,9 mg/mL) e 2 (CL 50 = 29,8 mg/mL) foram citotóxicas contra A. salina, enquanto que a fração aquosa (FA, CL 50 >> 1000 mg/mL) mostrou-se inativa. Conclui-se que a citotoxidade observada para HME e FD pode ser atribuída à presença dos ácidos caurenóico (1) e grandiflorênico (2) nestes extratos.Unitermos: Wedelia paludosa, Asteraceae, atividade citotóxica, toxidade em Artemia salina, ácido caurenóico, ácido grandiflorênico, CLAE.ABSTRACT: Wedelia paludosa D.C. (Asteraceae) is an ornamental species occurring in many regions of Brazil. Aiming to find new cytotoxic compounds, the hydromethanol extract of W. paludosa (HME), as well as the dichloromethane (DF) and water (WF) fractions resulting from its partition, were submitted to the brine shrimp lethality bioassay (BSLB) in order to evaluate their cytotoxicity. Dichloromethane fraction (DF) was shown to be the most cytotoxic fraction (LC 50 = 140.6 mg/mL), and its analysis by reversed phase high performance liquid chromatography (RP-HPLC) revealed ent-kaurenoic (1, 6.22 ± 0.23%) and grandiflorenic (2, 3.22 ± 0.31%) acids as important constituents. HME (LC 50 = 980 mg/mL), DF (LC 50 = 140.6 mg/mL), 1 (LC 50 = 15.9 mg/ mL) and 2 (LC 50 = 29.8 mg/mL) were found to be cytotoxic, while the water fraction (WF, LC 50 >> 1000 mg/mL) was inactive. As conclusion, the cytotoxicity observed for HME and DF is mainly due to the presence of 1 and 2 in their constitution.

Natural products are a very productive source of leads for the development of medicines. Seven Brazilian Asteraceae adult plants were randomly chosen. The current study was designed to evaluate the antiprotozoal and cytotoxic activities in vitro of 21 extracts obtained. Phytochemical properties of the most active extracts also were checked. Cytotoxic activity was evaluated by 3-(4,5-dimethylthiazol-2-yl)-2,5-diphenyltetrazolium bromide (MTT) method against human tumor cell lines (HCT-116, OVCAR 8 and SF-295). The antiprotozoal activity was evaluated against Trichomonas vaginalis, Entamoeba histolytica and Giardia lamblia. None of the extracts showed antiprotozoal activity. However, 76% of the extracts displayed moderate to high in vitro cytotoxic activities against human cancer cell which suggests that they are a promising source of anticancer compound, since none of the extracts showed hemolytic activity. Terpenoids, flavonoids, saponins, tannins, besides other compound classes, were identified and may be responsible for their antitumor activity. Cytotoxic assays indicate the anticancer potential of Asteraceae species from Brazil.

RESUMO:No Brasil as espécies de Mikania glomerata Spreng. e M. laevigata Sch. Bip. ex Baker, Asteraceae, popularmente conhecidas como guaco, são amplamente utilizadas com evidências pré-clínicas para o tratamento de doenças onde estejam envolvidas suas ações broncodilatadoras, antialérgicas, antiasmáticas, anti-inflamatórias, antiulcerogênicas e relaxantes da musculatura lisa. Em virtude das propriedades terapêuticas atribuídas as essas espécies, ambas foram oficializadas na farmacopéia brasileira, e por serem bastante semelhantes, dificilmente são diferenciadas. Com o objetivo de contribuir com novas pesquisas, a presente revisão descreve, de forma sistematizada, sobre os aspectos farmacológicos, toxicológicos, químicos, agronômicos, morfoanatômicos, genéticos e o uso nos programas de fitoterapia do Brasil destas espécies. As principais diferenças e semelhanças entre as duas espécies são evidenciadas através da comparação das informações baseadas na literatura específica.Unitermos: Asteraceae, Mikania glomerata, Mikania laevigata, guaco, plantas medicinais.ABSTRACT: "Mikania glomerata Spreng. e M. laevigata Sch. In Brazil the species of Mikania glomerata Spreng. and M. laevigata Sch. Bip. ex Baker, Asteraceae popularly known as guaco, are widely used with preclinical evidence for the treatment of diseases whose actions involve bronchodilators, antiallergic, anti-asthmatic, anti-inflammatory, antiulcerogenic and smooth muscles relaxant. Due to the therapeutic properties attributed to these species, both were included in the official Brazilian Pharmacopoeia, and because of their similarity, they are hardly differentiated. In order to contribute to the new research, this review describes their several aspects: pharmacology, toxicology, chemical, agricultural, anatomical, genetics and the use in herbal medicine programs in Brazil, in a systematic way. The main differences and similarities between the two species are evidenced by comparing the information based on literature.
