Bei der Hydrolyse des Kondensationsproduktes aus Nicotinoylessigester, Formaldehyd und Acetaminomalonester entsteht 2-[P-Pyridyl]-A2-pyrrolincarbonsaure-(5) (I1 a), die in das Tabakrauch-Alkaloid Myosmin (VI) und in DL-Nicotin (Va) iibergefiihrt wird. Analog wirdausgehend von Isonicotinoylessigesterdas y-Isomere des Nicotins (V b) synthetisiert. Zwischenstufen der Synthese sind die substituierten Proline IIIa bzw. IIIb. Substituierte Acylaminomalonester sind Ausgangsverbindungen fur die Synthese von Aminosauren2.3). Verknupft man z. B. durch unsymmetrische Dreikohlenstoff-Kondensation Acetaminomalonester mit Formaldehyd und Malonester, so ergibt die Saurehydrolyse Glutaminsaure-hydrochlorid3). Die Kondensation fuhrt bei Verwendung von P-Ketocarbonsaureestern, @-Diketonen, @-Phenylketonen und P-Ketonitrilen anstelle des Malonesters zu y-Ketoacylaminomalonestern4,~), aus denen bei der sauren Hydrolyse in 2-Stellung substituierte A2-Pyrrolin-carbonsauren-(5) entstehen, die durch Hydrierung in 5-substituierte Proline iiberfuhrbar sind. So erhielten HELLMANN und AICHINGER~) durch Hydrolyse von [Dihydroresorcyl-(2)-methyl]acetaminomalonester in maDiger Ausbeute 5-Carboxy-A2-pyrrolin-buttersaure-(2). S A N N O~) gewann 5-Methyl-prolin durch Hydrolyse und Hydrierung des aus Acetylaceton und der MANNICH-Base von Acetaminomalonester gebildeten Kondensationsproduktes. Liebigs Ann. Chem. Bd. 672 7

Es wird eine Vorschrift gegeben für die Bereitung von 3‐Aminopyridin aus 3‐Brom‐pyridin in grösseren Mengen. Die Darstellung von N‐(3‐Pyridyl)‐pyrrol wird beschrieben: bei dieser Reaktion entsteht bis‐(3‐Pyridyl)‐Harnstoff als Nebenprodukt. 3‐(3′‐Pyridyl)‐pyrrol (3, 3′‐Nornicotyrin) wird durch Reduktion mit Zinkstaub und Salzsäure in 3‐(3′‐Pyridyl)‐pyrrolidin (3, 3′‐Nornicotin) umgesetzt; dieses Nornicotin ist bereits nach einer anderen Methode von Späth und Kainrath erhalten worden. Durch Erhitzen von 3, 3′‐Nornicotin mit Formaldehyd und Ameisensäure erhielten wir d, l‐3, 3′‐Nicotin, welche Base ebenfalls schon von Späth und Kainrath synthetisiert worden ist. Durch Erhitzen von 3‐(2′‐Pyridyl)‐pyrrol mit Zinkstaub und Salzsäure erhielten wir 3‐(2′‐Pyridyl)‐pyrrolidin (3, 2′‐Nornicotin). Diese Base wurde durch Erhitzen mit Formaldehyd und Ameisensäure in d, l‐2, 3′‐Nicotin übergeführt.
