Synthesis and characterization of novel poly(γ‐benzyl‐L‐glutamate) derivatives tailored for the preparation of nanoparticles of pharmaceutical interest

Abstract: Four poly(γ‐benzyl‐L‐glutamate) (PBLG) derivatives bearing at one end specific groups were synthesized by ring‐opening polymerization of the corresponding γ‐benzyl‐L‐glutamate N‐carboxyanhydride using different amine‐terminated initiators. These moieties were chosen to introduce, on demand, specific functionalities in nanoparticles of pharmaceutical interest. The PBLG and PBLG derivatives were characterized by 1H NMR, viscosimetry, Fourier transform infrared spectroscopy and differential scanning calorimetry. … Show more

“…[28][29][30] In addition to surface functionalization, other factors such as particle size, shape, surface charge, and surface functionality also affect the interactions between particles and cells. 31,32 In our study, iron oxide NP surface was modified with dextran as confirmed by FTIR analysis. Utilizing DIONPs, we observed that the size of NPs in culture media was greater than its primary size, which is most likely due to the formation of the protein corona on the NPs in the buffer medium.…”

“…Carboxylic acids from PGlu were in the protonated state as evidenced by the band at 1710 cm −1 . PBLG-b-PGlu copolymer presented an α-helix structure, as shown by the amide I amide II and amide III bands at~1655 cm −1 ,~1550 cm −1 and~1260 cm −1 , respectively and the ester C=O band at 1735 cm −1 (see SM) (19)(20)(21).…”

“…1), were prepared by a new nanoprecipitation method, modified from a previous one (19). The use of NaOH was essential for nanoparticle preparation since ionization of the carboxylate groups in the hydrophilic PGlu block was necessary to avoid nanoparticle aggregation (SM, figure S7).…”

“…On the other hand, it has been shown previously that various chemical moieties could be quite easily introduced in the structure of poly(g-benzyl L-glutamate) to form various copolymers (Martínez Barbosa et al, 2007).…”
