Skeletal deformities induced by the intraperitoneal administration of deoxynivalenol (vomitoxin) in mice

Abstract: The contamination of drinking water by organic acids, selenium deficiency and the ingestion of fungal mycotoxins are the three main aetiological factors in the development of Kashin-Beck disease. An avian tibial chondrodysplasia induced by mycotoxins has been reported. Deoxynivalenol (DON) is one of many mycotoxins produced by the most common contaminating species of fungi. The pattern of skeletal malformations induced by its administration intraperitoneally to pregnant mice is reported. Costo-vertebral segmen… Show more

“…It is known that a decrease in intestinal calcium absorption and hypocalcaemia with concomitant impairment in vitamin D metabolism occur in DON poisoning (8). At the 3.5 ppm (3.5 mg kg -1 ) DON concentration, increased liver weight and a decreased concentration of serum protein and albumin, as well as serum and phosphorus were observed in pigs (19).…”

“…Avian tibial chondrodysplasia induced by mycotoxins has been reported (14). Moreover, contamination of food with mycotoxins has been suggested as a causal agent of Kashin-Beck disease in a human endemic osteoarticular condition (8).…”

“…In addition, the roles of some environmental factors have been studied experimentally by medical authors [7,3,6,14]. There was nothing in the history of the five dogs in the present series that pointed to particular genetic or environmental factors that might have caused the anomalies or predisposed the subjects to them.…”

“…[7][8][9][10][11][12][13] Toxic effects by DON seem to vary with species, dose, route of exposure, gender, and ages of each animal models, but the main developmental effects appear to be significant resorption of mouse embryos, skeletal malformations and complete resorption of fetuses were observed with increasing doses of DON which reduced live fetus number per dam. 14,15) Moreover, the developmental hematopoietic progenitors have been well documented for their susceptibility to DON. B lymphocyte precursor cells, CD34ϩ progenitor cells, megakaryocytic progenitors and granulomonocytic progenitors are highly responsive to this toxin and are supposed to undergo apoptotic cell death as a result from ribotoxic stress responses.…”

“…Most commonly observed effect of DON during in utero exposure is the fetal skeletal abnormality. 14) In utero exposure also could preclude the chance of any immunoteratological manifestations associated with changes in the systemic immunity. 11) Toxicity in the embryonic hematopoietic development has been extensively investigated using the avian embryonic eggs.…”
