The development of recovery processes using aqueous two-phase systems (ATPS) has been limited by the extensive experimental work required to establish the optimal system parameters to ensure selective partitioning of the product of interest. Although using full factorial experiments has been demonstrated to be an effective strategy for the characterization of the partitioning behaviour of biological products in ATPS, this approach is characterized by its costly and time consuming nature. As an alternative, the use of a robotic-aided strategy has been proposed. However, the need for high cost equipment may limit the generic implementation of this strategy. Based on practical experience using ATPS, practical strategies for the predictive design of primary recovery processes using polymer-salt systems were derived. To evaluate the generic application of the proposed strategies, four experimental models (B-phycoerythrin, C-phycocyanin, double layered rotavirus-like particles and lutein) were selected. The application of these strategies resulted in the development of simplified recovery processes for the selected experimental models. The practical review presented is considered a relevant contribution that will provide general rules to facilitate the establishment of ATPS processes, particularly for new researchers in the field.

All commercial corn oil is obtained by pressing corn germ and/or extracting the germ with hexane. In the current study, six types of corn oil were prepared by extracting corn germ, corn fiber and ground corn, each with hexane or with ethanol. The levels of lutein, zeaxanthin and other carotenoids were quantitatively analyzed in the six corn oils. The levels of lutein + zeaxanthin in the oil ranged from 2.3 lg/g for hexane-extracted corn germ oil to 220.9 lg/g for ethanol-extracted ground corn oil. These results indicate that a diet that includes 30 g (*2 tbsp) per day of the unrefined corn oil obtained by extracting ground corn with ethanol would provide *6 mg of lutein + zeaxanthin, the daily dosage that is currently considered to be necessary to slow the progression of age-related macular degeneration.

This study aimed to identify and determine the carotenoids from green microalga, Scenedesmus protuberans using analytical techniques. Identification of carotenoids was realized by comparing their absorption and mass spectral data with those of reference standards available and reported values. Chromatographic data were then combined with the spectroscopic information. The separation of carotenoids was achieved by C 30 column and high-performance liquid chromatography-diode array detection was used for their determination. In the present work, the carotenoid content of S. protuberans was found to be 1.45 mg/g of violaxanthin, 2.47 mg/g of all-trans-lutein, 0.15 mg/g of all-trans-α-carotene, 0.55 mg/g of all-trans-β-carotene, and 0.20 mg/g of 9 or 9′-cis-β-carotene. Due to lack of their standards, the amount of all-trans-loroxanthin and cis-isomers of other carotenoids could not be quantified. In order to validate the method, Certified Reference Material (BCR 485-Mixed vegetables) was used. In conclusion, this study can serve as a reference for the analysis of carotenoids in other microalgae.
