Consumers' desires to either reduce the risk of or manage a specific health condition through improved diet have stimulated the research of agricultural products for their potential health beneficial components such as tocopherols and natural antioxidants. Soft wheat is one of the major crops in Maryland, with little information available about its potentially beneficial components. This study examined eight selected Maryland-grown soft wheat varieties or experimental lines for their potential beneficial components including tocopherols, carotenoids, total phenolics and phenolic acids and their antioxidant properties, including Fe(2+) chelating capacity and free radical scavenging activities against 2,2-diphenyl-1-picrylhydrazyl radical (DPPH(*) ), radical cation ABTS(*)(+), and oxygen radical (ORAC). The results showed that all tested soft wheat grain samples contained alpha-tocopherol, with a range of 3.4-10.1 microg/g. Lutein was the primary carotenoid present in the grain samples at a level of 0.82-1.14 microg/g, along with significant amounts of zeaxanthin and beta-carotene. Vanillic, syringic, p-coumaric, and ferulic acids were found in soluble free, soluble conjugated, and insoluble bound forms in the grain extracts, with ferulic acid as the predominant phenolic acid. The eight soft wheat varieties differed in their antioxidant properties. The tested wheat grain samples exhibited ED(50) values against DPPH(*) of 23-27 mg of grain equiv/mL, ORAC of 32.9-48 micromol of Trolox equiv (TE)/g, and ABTS(*)(+) scavenging capacity of 14.3-17.6 micromol of TE/g. These data suggest the possibility of producing soft wheat varieties rich in selected health beneficial factors for optimum human nutrition though breeding programs.

The retention mechanism and chromatographic behavior for different polar analytes under hydrophilic interaction chromatography (HILIC) conditions have been studied by application of different mobile phases and stationary phases to various analytes at different temperatures. In addition to the commonly accepted mechanism of analyte liquid-liquid partitioning between mobile phase and water-enriched solvent layer which is partially immobilized onto the surface of the stationary phase, hydrogen-bonding, hydrophobic interaction, and ion-exchange interactions may also be involved. The predominant retention mechanism in HILIC separation is not always easily predictable. It can depend not only on the characteristics of the analytes but also on the selection of mobile and stationary phase compositions. The objective of this review is to evaluate the potential application of column temperature and mobile phase composition toward improving HILIC selectivity. The functional groups from analyte structures, stationary phase materials and organic mobile phase solvents will be highlighted.

Cold-pressed onion, parsley, cardamom, mullein, roasted pumpkin, and milk thistle seed oils were characterized for their fatty acid (FA) composition, tocopherol content, carotenoid profile, total phenolic content (TPC), oxidative stability index (OSI), color, physical properties, and radical-scavenging capacities against peroxyl (oxygen radical-scavenging capacity) and stable DPPH (diphenylpicrylhydrazyl) radicals. Parsley seed oil had the highest oleic acid content, 81 g/100 g total FA, and the lowest saturated fat among the tested oils. Roasted pumpkin seed oil contained the highest level of total carotenoids, zeaxanthin, βcarotene, cryptoxanthin, and lutein at 71 µmol/kg and 28.5, 6.0, 4.9, and 0.3 mg/kg oil, respectively. Onion seed oil exhibited the highest levels of α-and total tocopherols under the experimental conditions. One of the parsley seed oils exhibited the strongest DPPH· scavenging capacity and the highest oxygen radical absorbance capacity (ORAC) value of 1098 µmol Trolox equiv/g oil. However, ORAC values of the tested seed oils were not necessarily correlated to their DPPH· scavenging capacities under the experimental conditions. The highest TPC of 3.4 mg gallic acid equiv/g oil was detected in one of the onion seed oils. The OSI values were 13.3, 16.9-31.4, 47.8, and 61.7 h for the milk thistle, onion, mullein, and roasted pumpkin seed oils, respectively. These data suggest that these seed oils may serve as dietary sources of special FA, tocopherols, carotenoids, phenolic compounds, and natural antioxidants.

Peptides abundant in food and protein hydrolysates are known to be important to process flavors. The present study reports the volatile profile of the Maillard reactions of glycine, diglycine, and triglycine. The reaction with glucose was conducted at 0-100% water content in glycerol medium at 160 degrees C for 1 h. Volatile compounds were quantified by stir bar sorptive extraction-gas chromatography-mass spectrometry, and nonvolatile compounds were quantified by high-performance liquid chromatography-tandem mass spectrometry. The major volatiles produced from each of the reaction systems were trimethylpyrazine and 2,5-dimethylpyrazine. Volatile generation increased as water decreased, and the overall reactivity of the glycine and glycine peptides in volatile formation was glycine approximately triglycine > diglycine. Triglycine was very unstable and mainly degraded into cyclic Gly-Gly and glycine, whereas diglycine had a higher stability than triglycine toward hydrolytic cleavage of the peptide bond. The amounts of glycine, diglycine, cyclic (Gly-Gly), and triglycine in the peptide-glucose reaction mixtures at different water content were reported.
