Simultaneous Estimation of Glimepiride, Rosiglitazone and Pioglitazone Hydrochloride in the Pharmaceutical Dosage Form

Abstract: A simple, specific, accurate and economical gradient reversed phase liquid chromatographic (RP-HPLC) method was developed and subsequently validated for the determination of glimipiride, rosiglitazone and pioglitazone hydrochloride. Separation was achieved with a nucleodur C-18 column having 250x4.6 mm i.d. with 5 µm particle size and water HPLC grade adjusted to pH 3.0 using diluted orthophosphoric acid and acetonitrile (80:20 v⁄v) with gradient program as eluent at a constant flow rate of 0.8 ml per min. “…There are some HPLC methods [17][18][19][20][21][22][23][24] reported in the literature, but these methods are developed only for individual analysis of Pioglitazone and Rosiglitazone in bulk and formulation studies. The developed HPLC method was utilized for the estimation of the combined drugs by in vitro method.…”

“…Although several analytical methods were reported for analysis of each component individually, few methods were reported for the simultaneous determination of MET and PIO in combination dosage forms. These methods include; spectroscopic and chemometric methods [7][8], thin layer chromatographic (TLC) methods [9] and high performance liquid chromatographic (HPLC) method [10][11][12][13][14][15][16]. On the other hand, the only report for their simultaneous determination in plasma sample was using LC-MS technique [17].…”
