Regulation of Catalase Activity in Mice of Different Ages

Abstract: The renewal of catalase activity in C57BL/6J male mice of different ages was followed for 24 h after the removal of activity with 3-amino-1,2,4,-triazole (AT). The return of activity after injection of AT in both liver and kidney of old animals was significantly slower than that of either young or middle-aged mice. Some old mice failed to renew any catalase activity following treatment with AT. This suggests either an increase in the rate of degradation, or decrease in the rate of synthesis, of catalase in old… Show more

“…Catalase activity was found to be higher in liver than in kidneys of mice [4] and rat [10], On the contrary, our results indicate that the enzyme activity is higher in kidneys than in liver of garden lizards. Earlier we reported a similar tissue difference in another anti-oxi dative enzyme, glucose-6-phosphate dehydro genase G6PDH [11], It is possible that as per physiological demands, the enzymatic anti oxidant defenses are more prominent in kid neys than in liver of this species of poikilothermic vertebrate.…”

“…The recovery of enzyme activity following inhibition of catalase in liver and kidney with a herbicide, aminotriazole, was found to be slower in old than in young and middle-aged mice [4], A similar decline in response of senescent fruit flies to renew catalase after aminotriazole inhibition has also been reported [12], While these re sults indicated conformational changes in cat alase of aged organisms, contradictory results were observed in enzyme induction. Clofibrate, an anti-hypercholesterolaemic drug, in duced a similar degree of 2.5-fold increase in hepatic catalase activity of C57BL/6J mice of all age groups, and there was no age-related lag in enzyme induction [21], This raised the question as to whether catalase molecules are altered during ageing.…”

“…4). This agrees well with previous findings [Baird and Samis, 1971] although hepatic catalase activity is generally lower in old mice (unpublished data) and old rats [Ross, 1959; H a in in g and L eg a n , 1973]. Treatment with CPIB resulted in a rapid rise in hepatic catalase acti vity in animals of all ages, which reached a maximal, twofold induction 4 days after the initiation of treatment (fig.…”

“…Most of the results are conflicting or equivocal. For example, in studies of the effect ofaging on liver antioxidative enzymes such as GSH-Px, SOD, and catalase in rodents, some have shown an age-associated increase,12,13) others have shown an age-associated decrease, [14][15][16][17] and still others have shown changes not related to age. 18 ,19) These conflicting reports prompted us to study the changes in GSH, GSH-Px, and other antioxidative factors during the life span of mice that can be caused as a model of aging.…”
