Oral administration of the non-steroidal anti-inflammatory drug, Diclofenac diethylamine (DDEA) is often associated with gastrointestinal ulcers, extensive first-pass hepatic metabolism, and gastric bleeding. As an alternative to oral administration, a transdermal drug delivery system (TDDS) of DDEA was developed for topical administration, to quantify diclofenac diethylamine in plasma of rabbits for this, a sensitive Reverse Phase-high performance liquid chromatography (RP-HPLC) method was developed to compare DDEA in marketed gel and optimized formulation, using Carbamazepine as Internal standard (IS). DDEA provoke on Hypersil RP C18 column (250 mm × 4.6 mm 5 μm) using a mobile phase mixture of potassium dihydrogen buffer pH 2.5 and acetonitrile in the ratio of 30:70v/v at an isocratic flow rate of 1mL/min. The retention time of DDEA was found to be 5.3min. The calibration curve was linear over the concentration range from 50-750ng/mL of DDEA. This method was accurate for quantitative estimation of the drug in marketed gel and optimized formulation. The main aim of study is to compare Pharmacokinetic profile of diclofenac diethylamine in pharmaceutical dosage forms (Marketed gel & formulated transdermal patches) using WinNonlin software version 8.1.

Aim and objective: Pollen is reported in flowers and leaves of various medicinal plants and showed its beneficial effect on human health. The objective of our study is to evaluate the antioxidant effect of flowers (Calendula officinalis and Dianthus chinensis) especially pollen protein and also determining its immunogenic effect against specific protein (typhoid vaccine) antigen in human whole blood samples. Methods: In this study, analytical studies were conducted for estimating the protein content using Lowry test and qualitatively determined through high performance thin layer chromatography (HPLC). In addition, immunological studies were also conducted using pollen protein for evaluating its antioxidant potential (using DPPH (2,2-diphenyl-1-picryl-hydrazyl-hydrate) free radical assay) and determined its immunogenic effect using typhoid vaccine antigen and determining its hemolytic effect in human whole blood samples. Results: This work demonstrated that pollen protein content (determined through lowry test) at higher concentrations exhibited a significant declining in antigen specific T cell proliferation using optimized dose of typhoid vaccine antigen in human whole blood samples. In addition, pollen protein showed antioxidant property as well and does not show any hemolytic effect. Conclusion: The current study showed that pollen protein from Calendula officinalis and Dianthus chinensis possesses antioxidant property and less immunogenic in terms of declining in antigen specific T cell proliferation rate.

Background: Chromatography is one of the important laboratory technique in which the components of a mixture are separated on an adsorbent in order to analyze, identify, purify and quantify a mixture. Thin Layer Chromatography (TLC)is used to support the identity of a compound in a mixture when the Rf of a compound is compared with the Rf of a known compound. High Performance Thin Layer Chromatography is a sophisticated and automated form of Thin Layer Chromatography (TLC). The procedure simultaneously processes the sample and standard that results in better analytical precision and accuracy at a faster pace. Pharmacological/ Toxicological action of Nux Vomica is because of its active principles present in the seeds namely strychnine, brucine etc. This research paper aims to corelate the active principles present in Nux Vomica with the toxicological action of the same. Materials and Methods: 1. Standard Nux Vomica mother tincture was tested for its alkaloid markers and its correlation with the toxicological action was studied. 2. Analysis of the mother tincture was done using High Performance Thin Layer Chromatography. 3. Stationary phase consisted of TLC Aluminium sheets with silica gel 60 F253 pre-coated layer (20cm x 10cm), thickness-0.2mm, no. of tracks-18, band length-6mm. 4. Mobile Phase consisted of Chloroform: Methanol (9.5:0.5). 5. The plate was developed in developing chamber and observed under U.V. Light. Results: Colours seen on the HPTLC Plates of samples are greenwhich corresponds to strychnine, dark blue which corresponds to brucine, orange to alkaloids fluorescent green to sterols and pink to fatty acids which are evident on the chromatogram. Conclusion: Therapeutic action of Nux Vomica as noted in Homoeopathic Materia Medica is because of the active principles like strychnine, brucine, alkaloids, sterols, fatty acids present in it which is evident from the chromatogram.
