COVID-19 caused by severe acute respiratory syndrome coronavirus 2 (SARS-CoV-2) primarily appeared in Wuhan, China, in December 2019. At present, no proper therapy and vaccinations are available for the disease, and it is increasing day by day with a high mortality rate. Pharmacophore based virtual screening of the selected natural product databases followed by Glide molecular docking and dynamics studies against SARS-CoV-2 main protease was investigated to identify potential ligands that may act as inhibitors. The molecules SN00293542 and SN00382835 revealed the highest docking score of 214.57 and 212.42 kcal/mol, respectively, when compared with the co-crystal ligands of PDB-6Y2F (O6K) and 6W63 (X77) of the SARS-CoV-2 M pro. To further validate the interactions of top scored molecules SN00293542 and SN00382835, molecular dynamics study of 100 ns was carried out. This indicated that the protein-ligand complex was stable throughout the simulation period, and minimal backbone fluctuations have ensued in the system. Post-MM-GBSA analysis of molecular dynamics data showed free binding energy-71.7004 1/2 7.98, 256.811/2 7.54 kcal/mol, respectively. The computational study identified several ligands that may act as potential inhibitors of SARS-CoV-2 M pro. The top-ranked molecules SN00293542, and SN00382835 occupied the active site of the target, the main protease like that of the co-crystal ligand. These molecules may emerge as a promising ligands against SARS-CoV-2 and thus needs further detailed investigations.

Enhalus acoroides (E. acoroides) is one of the most common species in seagrass meadows. Based on the application of allelochemicals from aquatic plants to inhibit harmful algal blooms (HABs), we used E. acoroides aqueous extract against harmful algae species Phaeocystis globosa (P. globosa). The results showed that E. acoroides aqueous extract could significantly inhibited the growth of P. globosa, decrease the chlorophyll-a content and photosynthetic efficiency (Fv/Fm) values of P. globosa, followed by vacuolization, plasmolysis, and the destruction of organelles. Twelve types of major chemical constituents were identified in E. acoroides aqueous extracts by ultraperformance liquid chromatography-high resolution mass spectrometry (UPLC-HRMS), including six flavonoids, two homocyclic peptides, two long-chain aliphatic amides, one tannin, and one nitrogen heterocyclic compound. Flavonoids were the characteristic chemical constituents of E. acoroides aqueous extract. Furthermore, the antialgal activity of luteolin-7-O-glucuronide (68.125 μg/mL in 8 g/L E. acoroides aqueous extract) was assessed. The EC50–96 h value was 34.29 μg/mL. In conclusion, the results revealed that luteolin 7-O-glucuronide was one of the antialgal compounds of E. acoroides aqueous extract, with potential application as novel algaecide.

New polymeric microspheres containing azomethine (1a-1c and 2a-2c) were synthesized by condensation to compare the enzymatic properties of the enzyme glucose oxidase (GOx) and to investigate antimutagenic and antimicrobial activities.The polymeric microspheres were characterized by elemental analysis, infrared spectra (FT-IR), proton nuclear magnetic resonance spectra, thermal gravimetric analysis, and scanning electron microscopy analysis. The catalytic activity of the glucose oxidase enzyme follows Michaelis-Menten kinetics. Influence of temperature, reusability, and storage capacity of the free and immobilized glucose oxidase enzyme were investigated. It is determined that immobilized enzymes exhibit good storage stability and reusability. After immobilization of GOx in polymeric supports, the thermal stability of the enzyme increased and the maximum reaction rate (V max ) decreased. The activity of the immobilized enzymes was preserved even after 5 months. The antibacterial and antifungal activity of the polymeric microspheres were evaluated by well-diffusion method against some selected pathogenic microorganisms. The antimutagenic properties of all compounds were also examined against sodium azide in human lymphocyte cells by micronuclei and sister chromatid exchange tests.
