Genetic diversity analysis is the first step to determine the level of genetic diversity of kemiri sunan accessions. High genetic diversity in generative propagated germplasm collection is a basic foundation to develop new high-yielding varieties. The research aimed to detect the genetic diversity of kemiri sunan population in NTT based on RAPD markers. The study was conducted at the Integrated Laboratory, Balittri, Sukabumi from May to July 2019. A total of 32 kemiri sunan accessions were obtained from a private plantation owned by PT. BHLI in Bajawa, Flores, NTT analyzed its genetic diversity using 40 RAPD markers. Markers that produced polymorphic bands were scored in binary data format and were then used for genetic diversity analysis using the NTSYS program. Results showed that 11 RAPD markers were polymorphic and generated a total of 41 bands consisting of 32 polymorphic bands (78.05%) and 9 monomorphic bands (21.95%). The genetic diversity analysis with a genetic similarity value of 0.737 showed that the 32 kemiri sunan accessions were divided into 2 clusters. Among those, two accessions (T2 and T4) can be selected as candidates for new high-yielding varieties, because they are located in different groups and showed high per plant fruit number. The result of the study also obtained the genetic distance value between kemiri sunan accessions varies from 0.00 to 0.46. The combination of two kemiri sunan accessions with a high genetic distance value should be useful as parents to obtain F1 progeny with a high heterosis effect.

Information about the genetic diversity of Red wood is needed to improve the quality and quantity of the plant. This information can be provided by analyzing plant DNA using molecular markers which are polymorphic and generate clear and thick bands. The objective of the study was to obtain primers that could generate clear and polymorphically bands from Red wood DNA. The evaluated Red wood was from Alor, Sika, Lembata, Rote, and Timur Tengah Selatan (TTS) provenances. As many as 54 primers screened to polymorphism assay. Those primers consisted of RAPD, SSR, and chloroplast.The SSR primers were developed from Pterocarpus indicus (six primers) and Prosopis alba (12 primers). Chloroplast primers were divided into two types, four Rubisco Large Sub-unit genes (rbcL) and 12 MaturaseK genes (matK). SSR primers of P. alba were not able to amplify Red wood DNA, whereas those of P. indicus could amplify the DNA. Three of each chloroplast primers successfully amplified the DNA, yet, each primer could only amplify one to four of 12 evaluated samples. rbcL 2 generated the highest number of sample, and samples from Lembata were amplified the most. Moreover, 16 RAPD primers were able to generate DNA.

The information on diversity needs to be disclosed as a basis for consideration in the use of genetic resources. The objective of this study is to reveal the diversity and the classification of tea germplasm based on morpho-agronomic characteristics. The experiment was carried out from June to December 2018 in Gunung Putri Experimental Garden, Cianjur, West Java, Indonesia. The materials of the study used were 13 first generation tea clones. The research used was the observation method.s. The level of kinship among the clones was calculated using cluster analysis with the PBSTAT program. The results of observations showed that there was a diversity of leaf margin characteristics, leaf shape, mature leaf color, and immature leaf color. The high diversity was seen in pecco+2 weight, pecco+3 weight, and petiole length. The highest weight of pecco+3 was found in clone Cin 1. The coefficient of dissimilarity of 0.34 was divided into 2 main groups. Group I consisted of the clone Cin 1 and the clone Cin 28. The second group consisted of 11 tea clones. The clone Cin 1 and the clone Cin 28 and the clone Mal 17 are the tea clones with the smallest similarity values.
