Polimixina B: efeito dose e tempo dependente na nefrotoxicidade in vitro

Abstract: Objective: To characterize the toxicity of polymyxin B (PmxB) in renal cell in different dosage and times. Methods: LLC-PK1 cells grown in 12 well multiwell plates were divided into the following groups: Control (CTL) -cells maintained in DMEM supplemented with 5%; G1 -cells exposed to concentration of 75µM PmxB G2 -cells exposed to concentration of 375µM PmxB. Each group was assessed at 24,48 and 72 hours as for cell viability (Acridine orange/ethidium bromide) and apoptosis (Hoechst 33342).Results: The data … Show more

“…Polymyxin B affects the osmotic properties and transport mechanisms of the cell membrane, leading to cell death. This action is not selective; polymyxin B also affects human cells due to similarities in membrane composition with bacteria (NEIVA et al, 2013). Another observation that seems to corroborate these results is the fact that the acidic environment induced by Otosporin ® interferes with cell metabolism, which requires a neutral pH for normal function.…”

“…Outros efeitos descritos são alergias levando à lesão cutânea (tipo urticária), dor no local de injeção (intramuscular), tromboflebite (injeção EV), febre e eosinofilia. As informações referentes ao uso em humanos durante a gravidez são limitadas, não sendo encontradas evidências de teratogenicidade humana com uso de polimixina B. O quadro neurotóxico reverte com a suspensão da droga (NEIVA et al, 2013).…”
