Assessment of intracanal medications cytotoxicity on L929 fibroblast cells

Abstract: ABSTRACT:This study aimed to evaluate the cytotoxicity of intracanal medications on L929 fibroblast cells at different periods of observation. The following experimental groups were studied: calcium hydroxide with camphorated paramonochlorophenol and glycerin (CPG); iodoform with glycerin (IG); calcium hydroxide with iodoform and distilled water (CIW); iodoform with distilled water (IW); calcium hydroxide with distilled water (CW); Otosporin ® (OT); and a control group composed of cells and culture medium. “…Lastly, after washing with phosphate-buffered saline (PBS), sample cells were lysed using 100 μL DMSO; the absorbance was measured at 540 nm with ELISA microplate reader (BioTek, Seattle, WA, USA). The cell viability was calculated using the percentage of untreated control cells [ 20 ].…”

“…In this study, the pulp fibroblasts were chosen once they grow easily and fast, and are located in areas susceptible to the effects of intracanal medications and their degradation products, as periapical tissues and periodontal ligaments. Also, these cells are the main producers of collagen tissue and therefore actively participate in the tissue repair process [17,18].…”

“…The use of biomaterials in the cicatricial process has drawn the attention of the scientific community, aiming to modulate or accelerate the process of tissue repair (FARIAS et al, 2016;CHEN;LIU, 2016;CARVALHO et al, 2018). Natural biomaterials such as medicinal plant substrates are considered a significant source of new chemicals with potential therapeutic effects MENESES, I. S. et al…”

“…2020 http://dx.doi.org/10.14393/BJ-v36n5a2020-50349 (CALIXTO, 2005;IPEK et al, 2005). In endodontics, factors such as cytotoxicity and resistance of microorganisms tend to replace the use of standard medications with those prepared from natural plants (KARKARE et al, 2015;DHARIWAL et al, 2016;FARIAS et al, 2016;TONEA et al, 2017). Among them, carvacrol (5isopropyl-2-methylphenol) -a monoterpene phenol present in the essential oils of numerous aromatic plants -has shown antimicrobial (KLEIN et al, 2013), antioxidant (BEENA et al, 2013), antinociceptive (MELO et al, 2012), and antiinflammatory effects (LANDA et al, 2009).…”
