Optimizing the gene transformation factors can be considered as the first and foremost step in successful genetic engineering and genome editing studies. However, it is usually difficult to achieve an optimized gene transformation protocol due to the cost and time-consuming as well as the complexity of this process. Therefore, it is necessary to use a novel computational approach such as machine learning models for analyzing gene transformation data. In the current study, three individual machine learning models including Multi-Layer Perceptron (MLP), Adaptive Neuro-Fuzzy Inference System (ANFIS), and Radial Basis Function (RBF) were developed for forecasting Agrobacterium-mediated gene transformation in chrysanthemum based on eleven input variables including Agrobacterium strain, optical density (OD), co-culture period (CCP), and different antibiotics including kanamycin (K), vancomycin (VA), cefotaxime (CF), hygromycin (H), carbenicillin (CA), geneticin (G), ticarcillin (TI), and paromomycin (P). Consequently, best-obtained results were used in the fusion process by bagging method. Results showed that ensemble model with the highest R 2 (0.83) had superb performance in comparison with all other individual models (MLP:063, RBF:0.69, and ANFIS: 0.74) in the validation set. Also, ensemble model was linked to Fruit fly optimization algorithm (FOA) for optimizing gene transformation, and the results showed that the maximum gene transformation efficiency (37.54%) can be achieved from EHA105 strain with 0.9 OD 600 , for 3.8 days CCP, 46.43 mg/l P, 9.54 mg/l K, 18.62 mg/l H, and 4.79 mg/l G as selection antibiotics and 109.74 μg/ml VA, 287.63 μg/ml CF, 334.07 μg/ml CA and 87.36 μg/ml TI as antibiotics in the selection medium. Moreover, sensitivity analysis demonstrated that input variables have a different degree of importance in gene transformation system in the order of Agrobacterium strain > CCP > K > CF > VA > P > OD > CA > H > TI > G. Generally, the developed hybrid model in this study (ensemble model-FOA) can be employed as an accurate and reliable approach in future genetic engineering and genome editing studies.

Wild Crambe species have greater potential than Crambe hispanica in industry, medicine, and as a vegetable, etc. A total of 53 germplasm accessions, belonging to 7 taxa, were collected from Turkey's natural flora. The accessions consisted of C. orientalis var. orientalis (18 accessions), C. orientalis var. dasycarpa (1 accession), C. orientalis var. sulphurea (2 accessions), C. tataria var. aspera (3 accessions), C. tataria var. tataria (26 accessions), C. grandiflora (1 accession), C. orientalis var. sulphurea, and C. maritima (2 accessions). In this study, the seed fatty acid compositions and oil contents were determined, and the data were used for taxonomic cluster, correlation, and principal component analyses. Important correlations were determined among the fatty acids; however, the oil contents were not correlated. Altitude was positively correlated with linolenic acid, while negatively correlated with oleic and linoleic acid. For the principal component and correlation analyses, 7 major fatty acids (>1%) were used, including palmitic (C16:0), oleic (C18:1), linoleic (C18:2), cis-11 eicosenoic (20:1), linolenic (C18:3), erucic (C22:1), and nervonic acid (C24:1). A total of 17 fatty acids were used for the cluster analyses. Two major clusters were formed, where the first consisted of C. orientalis, C. tataria, and C. grandiflora taxa, while the second consisted 3 of only C. maritima taxa. The dendrogram based on the fatty acid values clearly discriminated the species groups; however C. tataria was not located close to C. maritima, contrary to previous molecular cluster studies.

Sustainable Cornelian Cherry Production in Montenegro: Importance of Local Genetic Resources

Cornelian cherries has been grown Balkan peninsula no apparent disease and pest problems for centuries. The most important pomological and technological properties of cornelian cherry genetic resources (eleven local and introduced varieties and selections) in Gornje Polimlje, Montenegro were studied in four–year periods. Fruit weight, stone weight, and mesocarp/stone ratio shows great variation and varied between 1.89 to 6.16 g, 0.32 to 0.64 g and between 76.66 and 90.59%, respectively. Genotypes significantly differed each other in terms of yield determined as per trunk cross section area (TCSA). For better visualization of the agronomical traits important to the yield, i.e., yield per TCSA and fruit weight data is presented in combination as measured in the years of study. The best promising genotypes are Vladimirskiy, Chisinau yellow, and Krupnoplodni NS, which had consistently higher yield and large fruits for sustainable fruit production. Dry matter, total sugars, reducing sugars, sucrose, total acidity, Ca–pectate, vitamin C, total anthocyanins, total polyphenols, and ash content of cornelian cherry cultivars and selections were found between 19.81–24.46%, 10.94–14.47%, 10.44–13.09%, 0.24–1.23%, 1.92–2.51%, 2.76–4.43%, 41.09–101.27 mg/100 g, 0–157.06 mg/100 g, 183.91–307.9 mg/100 g, and 0.89–1.16%, respectively. The amount of potassium, which predominates in percentage of minerals in the ash, ranged from 2888 to 3574 mg/kg. The extensiveness of the study leads, therefore, to several clear conclusions and recommendations. Consistently, the best balance of biochemical elements in combination with good yield and fruit size measurements is found in selection Krupnoplodi NS. If consider fruit size and yield efficiency are priority, Vladimirskiy, Chisinau yellow, Kosten 3, and Krupnoplodni NS have clear advantage over the other genotypes. The study highlights the importance of local cornelian cherry selections for sustainable cornelian cherry production in Montenegro.
