Osteogenic differentiation of human bone marrow mesenchymal stem cells in hydrogel containing nacre powder

Abstract: Nacre (or mother of pearl) can facilitate bone cell differentiation and can speed up their mineralization. Here we report on the capability of nacre to induce differentiation of human bone marrow mesenchymal stem cells (hBM-MSCs) and the production of extracellular matrix. hBM-MSCs were encapsulated in an alginate hydrogel containing different concentrations of powdered nacre and cultured in the same environment until Day 28. Analysis of osteogenic gene expression, histochemistry, second harmonic generation (S… Show more

“…These two types of cells could be seeded in a composite scaffold enriched with nacre. Flausse et al have demonstrated the efficiency of powdered nacre in stimulation of differentiation and bone matrix production of MSCs seeded in alginate hydrogel. Powdered nacre was still used in an injectable form (mixed with venous blood) to fill bone defects in vertebral and maxillar sites of sheep .…”

“…In a recent study antioxidant peptides from P. fucata were found to inhibit the photoaging effect in mice [64]. In addition, enhanced osteogenic differentiation in hydrogel containing nacre powder suggested possible use of nacre in various future tissue-engineering applications [65]. Furthermore, the diversity of shell matrix proteins may be implicated in various biological activities [66].…”

“…fabricated Alg beads with nacre (mother of pearl) powder and culture them with BMSC cells. Observation of histochemistry suggested the formation of a new tissue composed of collagen fibers in the presence of nacre and HAp crystals in the Alg beads . Cell attachment and mineral deposition were enhanced when the sphere containing β‐TCP supplemented with silica and silicatein into PLGA MS. Mineralization increased from 0.29 ± 0.06 in β‐TCP micro to 0.51 ± 0.09 in addition to silica.…”
