Novel Competitive Chemiluminescence DNA Assay Based on Fe3O4@SiO2@Au-Functionalized Magnetic Nanoparticles for Sensitive Detection of p53 Tumor Suppressor Gene

Abstract: A simple, rapid response time and ultrahigh sensitive chemiluminescence (CL) DNA assay based on FeO@SiO@Au-functionalized magnetic nanoparticles (Au-MNPs) was developed for detection of p53 tumor suppressor gene. In this study, 2',6'-dimethylcarbonylphenyl-10-sulfopropyl acridinium-9-carboxylate 4'-NHS ester (NSP-DMAE-NHS), as a new kind of highly efficient luminescence reagent, was immobilized on the complementary sequence of the wild-type p53 (ssDNA) to improve the detection sensitivity. The optimal concentr… Show more

“…The sensing performance of Au|MPA- Cp53 Ab1 < Tp53 Ag > Pp53|CIS@ZnS‑NCs Ab2|Zn2+–AIE‑AuNCs for simultaneously determining CEA and p53 is identical to the single-analyze sensing performance of Au|MPA-Ab 1 <Ag>Ab 2 |Zn 2+ -AIE-AuNCs for determining CEA and Au|MPA-C p53 <T p53 > P p53 |CIS@ZnS-NCs for determining p53. When compared with the previously described methods for CEA or p53 determination (Table S2), the proposed approach for the simultaneous detection of both CEA and p53 can exhibit promising performance, which is comparable to, or even better than, that of previously reported approaches for single-component detection of CEA or p53. , …”

“…NSP‐DMAE‐NHS (NDN), a novel and effective luminescent reagent, is more efficient than classical chemiluminescent reagents such as luminol and shows higher performance in dilute alkaline hydrogen peroxide. [ 20 ] NDN displayed chemiluminescence profiles in the blue region (≈440 nm), which highly overlapped with the absorption of the as‐prepared nHOF (Figure 1e). The surface carboxyl groups of nHOF were activated and subsequently modified with H 2 N–PEG–NH 2 ; this enhanced its biocompatibility and favored NSP‐DMAE‐NHS loading.…”

“…P53 is a human tumor suppressor gene, which is of great significance to human health. Therefore, Wang [193] established a CL method to detect the gene and used a new type of efficient luminescent material, 2′,6′-dimethylcarbonylphenyl-10-sulfopropyl acridinium-9-carboxylate 4′-NHS ester (NSP-DMAE-NHS). The method comprises the following steps: firstly coating BSA on AuMNPs, and then connecting amino wildtype p53 genes.…”
