Myofibroblasts from salivary gland adenoid cystic carcinomas promote cancer invasion by expressing MMP2 and CXCL12

Abstract: This study demonstrated the presence of myofibroblasts in ACC. Myofibroblasts might be related to the aggressive growth behaviour of ACC, owing to their high levels of expression of MMP2 and CXCL12.

“…Our previous study demonstrated that ACC-derived CAFs promoted ACC cell invasion in a 3D matrix in a spheroid fashion[ 21 ]. Furthermore, we demonstrated that ACC-derived CAFs showed high levels of MMP2 and CXCL12 expression, which might be related to the aggressive growth behavior of ACC[ 22 ].…”

“…Expression of MMP-2, MMP-9 and their tissue inhibitors in benign and malignant salivary gland tumors showed an imbalance between MMPs and their tissue inhibitors in malignant neoplasms and invasive properties related to MMP-2 [15]. Guan et al, [10] also evaluated MMP-2 expression in salivary gland adenoid cystic carcinoma. Their result emphasized on the role of high levels of MMP-2 expression in aggressive behavior of tumor.…”

“…Extracellular matrix (ECM) molecules are modified by matrix metalloproteinases (MMPs), a family of enzymes that can modulate cell density by creating space for migration, releasing ECMbound growth factors and activating signaling molecules. MMP-2 and MMP-9 are type IV collagenases that degrade the major structural protein of extracellular matrix and basement membrane [10]. The prognostic significance of MMP-2, 9 expressions has been reported in prostate cancer and OSCC [11,12].…”

“…It shows a strong capacity for local invasion and a high incidence of distant metastasis Dantas et al 2015). Our previous studies demonstrated that carcinoma-associated fibroblast (CAF) derived from SACC showed high levels of MMP2 and CXCL12 expression, which might be related to the aggressive growth behavior of SACC (Guan et al 2015). Furthermore, we demonstrated that SACCderived CAF led SACC invasion by creating an invasive track in the extracellular matrix (ECM) (Li et al 2016).…”
