Establishment and characterization of a carcinoma-associated fibroblast cell line derived from a human salivary gland adenoid cystic carcinoma

Abstract: Salivary gland adenoid cystic carcinoma (SACC) is one of the most common malignancies in the oral and maxillofacial region. Carcinoma-associated fibroblast (CAF) is an important component in the tumor microenvironment and participates in SACC progression. In this study, we established a CAF cell line derived from a human SACC and named it CAF-SA. It was identified that CAF-SA expressed typical CAF biomarkers. Then, we studied the cellular communications between CAF-SA, tumor cells and endothelial cells. It was… Show more

“…Besides representing tissue structure, the parallel design is beneficial to mimic pathological processes in vitro, like the invasion and metastasis of adenoid cystic carcinoma (ACC) ( Liu et al 2010 ; Kong et al 2016 ; Kong et al 2018 ). ACC-on-a-chip was built to investigate the invasion pattern of salivary gland ACC ( Li et al 2016 ) ( Fig.…”

“…Thus, spheroid culture is expected to be a milestone in the advancement of cancer research mainly due to the advantage of multicellular cancer spheroid culture (Tanaka and Mishima, 2020). Kong et al (2018), established a human SACCderived CAF cell line (CAF-SA). Additionally, using a wound-healing assay, the group evidenced that CAF-SA significantly promoted migration ability, invasion in SACC-83 and SACC-LM cell lines, and increased angiogenic capacity of endothelial cells.…”

“…Another factor that contributes to metastasis is the presence of cancerassociated fibroblasts (CAFs) in the tumor stroma. Cells derived from ACC associated with CAFs were transplanted subcutaneously in nude mice, forming tumors with greater volume than those transplanted without CAFs, which was accompanied by a higher positivity for Ki-67 in tumors cells (Kong et al, 2018).…”
