Monoterpene Hydroperoxides with Trypanocidal Activity from Chenopodium ambrosioides

Abstract: Four monoterpene hydroperoxides were isolated from aerial parts of Chenopodium ambrosioides along with ascaridole (1), the anthelmintic principle of this plant, as anti-trypanosomal compounds. The structures of these monoterpenes were determined to be (-)-(2S,4S)- and (-)-(2R,4S)-p-mentha-1(7),8-dien-2-hydroperoxide (2a and 3a) and (-)-(1R,4S)- and (-)-(1S,4S)-p-mentha-2,8-dien-1-hydroperoxide (4a and 5a) on the basis of spectroscopic methods and chemical correlations. In vitro trypanocidal activities of ascar… Show more

“…Ascaridole has been documented with sedative and pain-relieving properties as well as antifungal eff ects 20 . Ascaridole was found to be a potent inhibitor in vitro development of Plasmodium falciparum 21 , Trypanosoma cruzi 22 , and Leishmania amazonensis 23 . Ascaridole also showed activity against diff erent tumor cell lines in vitro (CCRF-CEM, HL60, MDA-MB-231).…”

“…The activities were stronger than that of the anthelmintic monoterpene endoperoxide, ascaridole (MLCϭ23 mM) and were comparable to those of monoterpene hydroperoxides isolated from Chenopodium ambrosioides. 8) In the HeLa cell infection assay, compound 3 very strongly inhibited the infection of the trypomastigotes at 1 mg/ml (Table 1). Dehydrocostus lactone (1) also significantly inhibited the infection at 1 mg/ml, whereas zaluzanin D (2) did not inhibit the infection at the same concentration.…”

“…The bicyclic monoterpene ascaridole and the phenylpropanoid safrole (Fig. 4) can be found in C. ambrosioides essential oil (Kiuchi et al, 2002;Tucker and Maciarello, 1998). This is an essential oil with anthelmintic activity that has been used to treat parasitosis and leishmaniasis (Monzote et al, 2009).…”
