Micronucleus assay in vivo provides significant prognostic information in human cervical carcinoma; the updated analysis

Abstract: A high frequency of micronuclei before radiotherapy and a slight increase of micronucleus frequency during radiotherapy measured after 10 fractions of 2 Gy were independent on stage, statistically significant adverse predictors of clinical outcome in cervical carcinoma patients treated with radiotherapy.

“…In the cervix tumor cells MN have good predictive value after one week of therapy -high number of MN compared to the background level predict good response to radiotherapy [29]. The same results were obtained by the research group of Wiedel -they investigated the tumor cells obtained with biopsy instead of exfoliated tumor cells [3,4].…”

“…It is well established that radio and chemother apy widely used for treatment of cancer patients induce chromosomal aberrations and micronuclei (MN) both in tumor [1][2][3][4] and normal (healthy) cells [5,6]. Because of some technical difficulties (invasive procedure to obtain tumor cells during and after treatment) cytogenetic disturbances are mostly studied in lymphocytes [7,8].…”

“…There was a good number of cervical cells with both chromosome loss (centromere positive micronucleus) and breakage (centromere-lacking micronucleus) in the LSIL and HSIL categories. In fact, many researchers have observed the higher frequencies of micronucleated cells among cancer patients compared to healthy individuals [29,30]. The ease and low cost of the detection of MN may allow it as a prognostic indicator during the planning and validation of programs for cancer monitoring and prevention.…”

“…The MN frequency in control (non-irradiated) cells, representing so-called spontaneous MN was regarded as a baseline level. The percentage of binucleated cells (%BNC) in non-irradiated cells was used for measurement of proliferation fraction in vitro (Widel et al, 2001). …”

“…Micronuclei can be measured after DNA staining by the Feulgen reaction, Giemsa or fluorescence dyes. Evaluation of micronucleus frequency is widely used for determination of cellular radiosensitivity (Wandl et al, 1989;Shibamoto et al, 1991, Widel et al, 2001. Generally, radiosensitive cells are more susceptible to radiation-induced micronucleus formation.…”
