The fumonisins, a family of food-borne carcinogenic mycotoxins, were first isolated in 1988 (21) from cultures of Fusarium verticillioides (Sacc.) Nirenberg (previously known as Fusarium moniliforme Sheldon). During the same year, the structures of the fumonisins were elucidated (6) and fumonisin B 1 was shown to cause equine leukoencephalomalacia (34). There have been numerous publications dealing with this group of novel, carcinogenic mycotoxins, and comprehensive reviews of different aspects of the fumonisins are available (20,22,23,24,35,36,37,41,43,46,52,55,60,61,66). Due to the widespread occurrence of the fumonisins in maize, a dietary staple in many countries, the carcinogenic risk of fumonisins to humans was evaluated by the International Agency for Research on Cancer in 1993, and the toxins produced by F. moniliforme were evaluated as "Group 2B carcinogens," i.e., probably carcinogenic to humans (24). This review focuses on the Fusarium species that produce fumonisins and the fumonisin analogs produced by each of these species.

RESUMO.-A fumonisina B 1 (FB 1 ) é um metabólito secundário produzido principalmente por Fusarium verticilioides em diversos tipos de alimentos, principalmente o milho, o qual constitui a base para composição de rações para várias espécies de animais domésticos. A FB 1 é particularmente tóxica para suínos, cujas manifestações clínicas são evidentes em animais expostos a altas concentrações de FB 1 na ração (em geral, acima de 30mg/kg). No entanto, são escassos os estudos sobre os efeitos da FB 1 em suínos alimentados com rações contendo baixas concentrações de fumonisinas, as quais são mais prováveis de serem encontradas em condições de campo. O objetivo do estudo foi avaliar os efeitos da exposição de leitões a baixos níveis de FB 1 na ra- ) is a secondary metabolite produced mainly by Fusarium verticilioides in several types of foods, particularly corn, which is the basis for composition of feed for several domestic animals. FB 1 is particularly toxic to pigs, being the clinical manifestations evident in animals exposed to high concentrations of FB 1 in the diet (generally above 30mg/kg). However, there are few studies on the effects of FB 1 on pigs fed rations containing low concentrations of fumonisin, which are most probably found under field conditions. The aim of the study was to evaluate the effects of a 28-day exposure of piglets to low levels of FB 1 in the feed on the weight gain, feed consumption, organ weights and histological aspects of the spleen, liver, lungs, kidneys and heart. Twenty-four pigs were assigned into 4 experimental groups and fed diets containing 0mg (control), 3.0mg, 6.0mg or 9.0mg FB 1 /kg diet. The different diets did not affect (P>0.05) the weight gain or the weight of organs examined. There were no macroscopic or histological lesions in the spleen, liver, kidneys and heart. However, histological lesions were found in the lungs from all animals fed rations containing fumonisin, hence indicating that none of the FB 1 levels used in the experiment could be considered as safe for piglets. Further studies on the mechanisms of toxic action of FB 1 in pigs are needed, particularly under conditions of prolonged exposure to low contamination levels in the diet.INDEX TERMS: Fumonisin B 1 , FB 1 , low levels, swine, toxic effects, mycotoxicosis.

Fumonisin B1 and ochratoxin A in beers made in Brazil

Samples of beer made in Brazil were analyzed for the presence of fumonisin B 1 (FB 1 ) and ochratoxin A (OTA). FB 1 was searched for in 58 beer samples from 30 plants located in nine states. The samples were concentrated and cleaned up with strong ion exchange column, derivatized with OPA and analyzed by HPLC with fluorescence detection. The limit of detection was 0.26 ng.mL -1 and the average recovery was 98%. Twenty-five samples contained FB 1 ranging from 1 to 40 ng.mL -1 . Beer (123 samples) from 36 plants located in 5 states were analyzed for OTA by means of immunoaffinity column cleanup followed by liquid chromatography associated with fluorescence. The detection limit was 0.1 ng.mL -1 and the average recovery was 92%. Five samples contained OTA in concentrations from 1 to 18 ng.mL -1 . The results indicate that FB 1 and OTA contamination in Brazilian beer is not geographically limited and that beer does not contribute significantly to FB 1 intake by consumers. In the case of regular high ingestion, beer could contribute sizably to OTA, intake although still below the maximum considered tolerable for the toxin. FB 1 foi pesquisada em 58 amostras de cerveja provenientes de 30 fábricas localizadas em nove Estados. As amostras foram concentradas, e a toxina isolada através de coluna de troca iônica forte, derivação com OPA e análise por CLAE com fluorescência associada. O limite de detecção foi 0,26 ng.mL -1 e a recuperação média foi de 98%. Vinte e cinco amostras continham FB 1 em concentrações de 1 a 40 ng.mL -1 . Cerveja (123 amostras) proveniente de 36 fábricas localizadas em 5 Estados foi analisada para OTA através de coluna de imunoafinidade seguida de CLAE com detector de fluorescência. O limite de detecção foi 0,1 ng.mL -1 e a recuperação média foi de 92%. Cinco amostras continham OTA em concentrações de 1 a 18 ng.mL -1 . Os resultados indicam que a contaminação da cerveja brasileira por FB 1 e por OTA não é geograficamente limitada e que não contribui significativamente para a ingestão de FB 1 por consumidores. Por outro lado, no caso de consumo alto e regular, esta pode contribuir substancialmente na ingestão de OTA, porém ainda abaixo do máximo considerado tolerável para a toxina.
