Isolation and Typing of Canine Parvovirus in CRFK Cell Line in Puducherry, South India

Abstract: A total of 128 faecal samples/rectal swabs were collected from dogs showing signs of diarrhea/enteritis in and around Puducherry, South India. Eighteen clinical samples, showing high HA titre of 1:512 and above and positivity by polymerase chain reaction (PCR) with CPV2ab primers, were subjected to virus isolation in CRFK cell line. Of the 18 samples processed, 3 samples (16.6%) were positive for CPV and were confirmed by haemagglutination, dot-ELISA, and IFAT. The three cell culture isolates were characterize… Show more

“…The virus-containing supernatants were used for DNA isolation or stored at –20 °C until further analysis. An HA test and TCID 50 test were used to characterize the viral isolates [ 22 , 23 ] . SCDN (Strain that isolated from Bulldog in Sichuan province) (CPV-2a) strain used as positive control [ 39 ].…”

“…Age-wise prevalence was found to be more between the age group of 3-6 months (below 6 months), followed by age group of 1-3 months, 6-12 months and the least above 12 months. This higher prevalence rate below 6 months also reported earlier [ 8 , 17 - 19 ]. The higher incidence of CPV below 6 months might be due to the affinity of the virus for rapidly multiplying intestinal crypt cells in weaning pups with higher mitotic index due to changes in bacterial flora as well as in the diet due to weaning [ 20 , 21 ].…”

“…In addition, our study showed that CPV isolation is feasible when faeces are processed shortly after collection. This procedure allows for enriching viral preparations for several purposes, including biological studies of viral replication, drug susceptibility and virus-host interactions ( Decaro and Buonavoglia, 2012 ; Parthiban et al, 2011 ). In a recent study, rural dogs living in households around six Atlantic forests in southeastern Brazil were serologically tested for CPV antibodies and 97% were positive ( Curi et al, 2016 ).…”
