In Vitro Determination of 6PGD Enzyme Activity Purified from Lake Van Fish (Chalcalburnus tarichii Pallas, 1811) Liver Exposed to Pesticides

Abstract: In the present study, the effect of methidathion, cypermethrin, and deltamethrin pesticides on Lake Van fish (Chalcalburnus tarichii Pallas, 1811) liver 6-phosphogluconate dehydrogenase enzyme activity was investigated due to the fact that these pesticides are extensively used to improve agricultural productivity in the Van region. 2',5'-ADP Sepharose 4B affinity chromatography was used to purify 6-phosphogluconate dehydrogenase enzyme from fish liver and SDS-PAGE technique was used to control the purity of th… Show more

“…The GR and G6PD activities decreased in all tissues, especially in Group C. There are a large number of studies focusing on the relationships between pesticides and antioxidant activities of GR and G6PD in different species (Güler et al, 2013;Lavarías and Garcıa, 2015). However, the number of the studies focusing on the oxidative stress caused by TE is limited.…”

“…[4,5] The detoxification reactions of these compounds have a complex process involving phase I, phase II xenobiotic metabolizing enzymes, and phase III transporters. [14] TA B L E 1 Summary of the GST purification procedure [7] They can catalyze the conjugation of the reduced glutathione (GSH) to electrophilic centers of many endogenous or exogenous toxic compounds, involving chemical carcinogens, oxidative stress products, insecticides, herbicides, and cancer chemotherapeutic agents.…”

“…The most important function of this enzyme is the production of ribose 5-phosphate and NADPH which are necessary for membrane lipids synthesis, reductive and nucleic acid synthesis (Kuo et al, 2000;Ceyhun et al, 2010). In addition, NADPH is a coenzyme which is involved in the synthesis of some amino acids, fatty acids and steroids, protecting the cells against the oxidant and detoxification of xenobiotics through the glutathione reductase peroxidase system (Ceyhun et al, 2010;Guler et al, 2013).…”
