Histochemical Study of Heparin‐positive Mast Cells in the Terminal Part of Porcine Ductus Choledochus and Papilla Duodeni Major

Abstract: The study presented in detail the localization and density of mast cells (MCs) in the intramural part of the common bile duct (CBD) and in the major duodenal papilla (MDP) of domestic swine. MCs' density (number/mm(2) ) in different layers of both of the duct and papilla was evaluated after toluidine blue staining. Their number was higher in the lamina propria mucosae than in the tunica muscularis of the studied structures. The localization of berberine-positive, (heparin containing) MCs and the ratio between … Show more

“…Mast cells exhibit metachromasia stained with toluidine blue due to granules in their cytoplasm (Bancroft & Gamble, ); Sun et al () in chicken proventriculus, Jing, Wang, and Chen () in the mammary glands of rat, Yıldız, Aydemir, Kum, and Eren () in quail caecum and Stefanov, Vodenicharov, Tsandev, and Sevrieva () in porcine common bile duct reported that mast cells were metachromatically stained with toluidine blue. While the sizes of the mast cells vary based on the tissue they are in, these generally round or oval cells may also be seen in the form of a spindle‐like shape depending on their location (Banks, ).…”

“…The majority of the histochemical methods for demonstration of mast cells depend on the content of heparin (6) and other glycosaminoglycans in their granules (7). Probably the most commonly employed one is the Toluidine blue staining, which shows the mastocytic granules as metachromatic structures (8), however producing a good amount of background staining of other cellular and extracellular elements (9).…”
