“…The mean value of motility obtained was higher than that of Kaka et al (2011) and Ansari et al (2014), each having a post thawing motility value of 43.25 ± 3.40% and 41.67 ± 2.90% in Kundhi buffalo. This difference was thought to be due to differences in buffalo breeds as the source of semen and genetic quality of each Bull.…”

“…The previous studies showed that the freeze-thaw process reduces the content of sperm glutathione in of boars [32], bulls [33] and goats [17]. And addition of glutathione to the freezing extenders improved the sperm quality [32, 34] though Marco-Jiménez et al (2006) [35] reported that GSH had no effect on rabbit sperm during cooling and freezing process. In this study, we further observed that the sperm glutathione decreased at each step, including cooling, equilibrium, freeze-thaw and post-thaw incubation.…”

“…Sperm DNA integrity was assessed by acridine orange assay (Ansari et al., ). The air‐dried smears of semen sample on glass slide were fixed overnight in Carnoy's solution (freshly prepared, methanol [Merck] and glacial acetic acid [Merck]; 3:1).…”

“…It is worth mentioning that the decision on the usability of any semen extender in routine artificial insemination programme is always based on the results of in vivo fertility rates of buffalo under field conditions (Akhter, Sajjad, Andrabi, Ullah, & Qayyum, ; Akhter, Ansari, et al., ; Akhter, Rakha, et al., ; Ansari, Rakha, Akhter, & Ashiq, ; Ansari et al., ). It was hypothesized that cryopreservation of buffalo semen in AndroMed ® extender may result in higher post‐thawed semen quality and in vivo fertility rate.…”
