An efficient and reproducible protocol for in vitro propagation of Dolichandra unguis-cati has been established for the first time from nodal segments. In order to enhance survival rate under ex vitro conditions, photosynthetic potential of in vitro grown plantlets was also studied through JIP test based analysis of polyphasic OJIP chlorophyll a fluorescence OJIP transients, density of active reaction centers, light harvesting efficiency, electron transfer rate, dissipation energy, maximum quantum yield of primary PSII photochemistry and photosynthetic performance index. The best morphogenetic in term of explants response (92.2 %), shoot number (3.43 AE 0.07) and shoot length (4.7 AE 0.31 cm) was obtained on Murashige and Skoog medium supplemented with 0.5 mg l À1 BAP and 1.0 mg l À1 TDZ. The shoots exhibited high frequency rhizogenesis on half strength medium augmented with 2.0 mg l À1 IAA. In vitro plantlets developed highest rate of photosynthesis on day 18 after the initiation of rhizogenesis. High survival rate (96.16%) under ex vitro conditions was observed when in vitro plantlets having high photosynthetic efficiency (Fv/Fm > 0.75) were subjected to hardening and acclimatization process. Plantlets with reduced photosynthetic performance exhibited low survival rate under natural conditions. The developed in vitro protocol will be useful for genetic improvement and multiplication of D. unguis-cati. The results of this study also show that photosynthetic screening of in vitro developed plantlets is highly essential after the rhizogenesis process to achieve higher survival rate under field conditions.

Antecedentes y Objetivos: Prosopis laevigata, especie de amplia distribución en México, es considerada un recurso con importancia ecológica y socioeconómica. A pesar de que sus semillas y frutos tienen valor nutrimental, los estudios sobre su variación morfológica y anatómica son escasos. Este trabajo es el primero en abordar la morfo-anatomía e histoquímica del fruto y semilla de P. laevigata y tiene como objetivo contribuir al conocimiento de su biología y proporcionar las bases para otras investigaciones con un enfoque ecofisiológico en esta y otras especies del género. Métodos: Muestras de frutos y semillas fueron recolectadas de árboles de P. laevigata en una localidad de Lagos de Moreno, Jalisco, México, para analizar sus caracteres morfométricos, anatómicos e histoquímicos. Adicionalmente, se estudió su composición micro elemental mediante microscopía electrónica de barrido (MEB) acoplada a espectroscopia de rayos X de energía dispersiva (EDS). Se aplicaron análisis ANOVA y de correlación para conocer la variabilidad intrapoblacional y asociación entre variables morfométricas. Resultados clave: El coeficiente de variación más alto (47.7%) fue para el número de semillas por fruto. El ANOVA demostró diferencias estadísticas significativas para las variables longitud (F=7.51, p<0.0001) y ancho de la semilla (F=8.75, p<0.0001); la correlación más alta (0.937) fue para ancho fruto-ancho semilla. Los frutos presentan epicarpo, mesocarpo con cristales de oxalato de calcio y endocarpo coriáceo. Las semillas tienen pleurograma, micrópilo subapical, testa gruesa con epidermis de macroesclereidas y osteoesclereidas en la hipodermis. Cotiledones contienen cuerpos de proteína y endospermo con almidón. La testa y el embrión presentan macroelementos (S, K, Mg) y microelementos (Al, Si, Cu). Conclusiones: La estructura morfológica, anatómica e histoquímica de frutos y semillas de P. laevigata fueron similares a otros Prosopis. La epidermis en empalizada, hipodermis con osteoesclereidas en la testa de la semilla, los estomas y cristales prismáticos en las vainas podrían representar caracteres ecofisiológicos importantes.

The irrational exploitation of native species of arid and semi-arid zones of Mexico has created the need to carry out research work that contributes to the reforestation of those zones, so the aim of this research was to establish the methodology of scarification and in vitro multiplication of the huizache. For scarification, three treatments were used; T1: Immersion in hot water for 20 minutes; T2: immersion in sulfuric acid for 1 h and cut with the scalpel before seeding; T3: sand the seeds and place them in water for 12 h. Once the scarification process was completed, the seeds were disinfected with 2% sodium hypochlorite and placed in flasks containing water-agar. For the multiplication phase, Murashige and Skoog medium supplemented with sucrose 30 g L-1, phytagel 2.5 g L-1 and four treatments were used: T1: BAP 0.5 mg L-1+ kin 0.5 mg L-1+ Chitosan 60 mg L -1; T2: BAP 1 mg L-1+ kin 0.5 mg L-1+ Chitosan 60 mg L-1; T3: BAP 0.5 mg L-1+ kin 0.5 mg L-1; T4: BAP 1 mg L-1+ kin 0.5 mg L-1. As results, in the scarification phase, the best treatments were T2 and T3 with 100% germination, and in the multiplication phase, T4 favored a multiplication coefficient of 3 and greater height and number of leaves in the explants than the rest of the treatments. These results will allow establishing the in vitro methodology of this important species.
