Approximately, 325 cultivars of bananas were found in Indonesia. They have variation in shape, taste, color of fruit pulp, essential vitamins and mineral. The β-carotene is one of essential nutrient in banana with various concentration in every cultivar. The aims of this research were to describe the morphological characteristics and to analyze β-carotene content of mature fruits of three different genomic groups of Indonesian dessert and cooking banana cultivars namely Berlin (AA/ dessert), Barangan (AAA/dessert), and Kepok (ABB/cooking). Morphological characterizations of fruits were conducted according to Descriptor for Banana from International Plant Genetic Resources Institute. The β-carotene content of mature banana pulp was analyzed using High-performance liquid chromatography (HPLC) method with three replications. Banana fruit characters were analyzed based on 15 descriptors. The bananas were then subjected to clustering analysis. The result showed that β-carotene content were 27 μg 100g -1 , 5 μg 100g -1 , and 3 μg 100g -1 fresh weight respectively. Berlin showed the highest value of β-carotene. Cluster analyses formed two groups. The first group consists of Berlin and Barangan while the second one consists of Kepok. This study can be used as a reference for people to consume bananas with high content of β-carotene in meeting the needs of vitamin A and as a consideration for the plant breeding especially for β-carotene improvement purpose.

Banana is one of the most essential commodities in Bali island. It is not only for nutrition sources but also for cultural and religious aspects. However, Bali banana genetic diversity has not been explored; therefore, in this study, we focused on its genetic relationship using a molecular approach. This research aimed to determine the genetic relationship of Bali banana cultivars using the internal transcribed spacer 2 (ITS-2) region as a molecular marker. A total of 39 banana samples (Musa spp. L.) were collected from Bali island. The ITS-2 DNA regions were then amplified and sequenced from both ends. ITS-2 sequences were predicted using the ITS2 Database (http://its2.bioapps.biozentrum.uni-wuerzburg.de/). The multiple sequences alignment was performed using ClustalX for nucleotide-based tree and LocARNA to provide the secondary structure information. Phylogenetic trees were constructed using neighbor-joining (Kimura-2-parameter model, 1,000 bootstrap). The result showed that two clades were formed, one clade was abundant in A genome (AA and AAA), and the other rich in the B genome (BB and ABB). This result suggested that cultivars that had similar genomic compositions tended to be grouped within the same clade and separated with different genomic compositions. This study gives perspectives that ITS-2 sequences in bananas are quite similar and differ much compared to other families. Secondary structure has been described to provide more robust resolving power in phylogenetic analysis.

Biodiversitas 23: 4124-4130. East Nusa Tenggara (NTT) is one of Indonesia's island that has many local maize cultivars. However, the existence of this local corn is threatened by the presence of high-yielding maize seeds. Therefore, characterization of local maize cultivars using molecular marker is needed to conserve the germplasm. This study aims to determine the genetic diversity of local maize cultivars (Zea maysL.) from East Nusa Tenggara based on ISSR markers. This research used eight local maize cultivars (Watar Ohaq, Watar Puluq, Wata Bura, Wata Kuma, Wata Bunga, Pena Muti, Boti Pulut, and Latun Wara). Young leaves of maize were used for DNA isolation. Total genomic DNA was isolated using Genomic DNA Mini Kit for Plant (Geneaid). DNA was then amplified using 5 ISSR molecular markers (ISSR-807, ISSR-808, UBC-809, UBC-811, UBC-834). DNA band profiles were scored based on the presence or absence of bands. Genetic variability was analyzed based on the UPGMA method using the MVSP ver application. 3.1. Local maize from NTT has high genetic diversity (similarity index between 0.29-0.65). Local maize cultivars were separated into two main clusters at a similarity index of 0.43 and the grouping pattern was not influenced by the geographic location of local maize origin.
