Bali Bananas (Musa spp. L.) Genetic Relationship Based on Internal Transcribed Spacer 2 (ITS-2)

Abstract: Banana is one of the most essential commodities in Bali island. It is not only for nutrition sources but also for cultural and religious aspects. However, Bali banana genetic diversity has not been explored; therefore, in this study, we focused on its genetic relationship using a molecular approach. This research aimed to determine the genetic relationship of Bali banana cultivars using the internal transcribed spacer 2 (ITS-2) region as a molecular marker. A total of 39 banana samples (Musa spp. L.) were coll… Show more

“…ITS2 primers were universal primers that could amplify ITS2 region because ITS2 is a molecular marker that was often used in plants because it had high variations to be able to differentiate the various banana cultivars. In addition, in previous study our group has also used the ITS2 primers to study various banana cultivars from Bali and other regions in Indonesia (Meitha et al 2020;Dwivany et al 2020). According to the literature, the size of ITS2 of bananas ranged from 205 to 227 bp, however, the results here were longer.…”

“…The parts of the Loka Pere plant that were sampled in this study were banana leaves and corms obtained from Adolang Village and Adolang Dhua Village, Pamboang District, Majene Regency, West Sulawesi. The DNA genome was extracted by using the CTAB method (Doyle 1990) [10] with a modification to scale down the volume. Each sample of banana leaf and sample of banana corms was ground using a mortar and pastel that had been given liquid nitrogen.…”

“…Based on these advantages, the use of the ITS area in phylogenetic analysis has the potential to be used in constructing relationships at lower taxonomic levels (groups and subgroups) of a plant [9] including the Loka Pere banana. ITS2 isolation for each type of banana had different treatments depending on the type of part of the banana plant to be DNA extracted [10]. It also depends on the DNA extraction method used [11].…”

“…Jenis genom tersebut berbeda dengan yang dinyatakan oleh Gusmiati et al, (2018). Dwivany et al, 2020 (Rinaldi et al, 2014), namun pengelompokkan kultivar pisang menggunakan karakter morfologi dianggap kurang akurat karena bersifat subyektif (Probojati et al, 2019). Klarifikasi tingkat ploidi dan jenis genom dapat dilakukan dengan menggunakan analisis kromosom (Simonikova et al, 2020).…”
