ABB), a unique local banana cultivar from Pasuruan, East Java, Indonesia. Biodiversitas 23: 3521-3532. Characterization of local banana (Musa spp.) germplasm is fundamental for identification and provides information on traits that support their sustainable conservation and optimum utilization. This study aims to characterize and discuss the potential and conservation of Pisang Kates,a local cultivar originatingfrom Tutur, Pasuruan, East Java, Indonesia. The observed characteristics include morphology, molecular, agronomy and yield performance, leaf anatomy and stomata, pollen morphology and viability, fruit total soluble solids and nutrients. Results showed that Pisang Kates hasa unique fruit shape ovoid to globose, weight of40-150 g, appear solitary and randomly attached directly to peduncle, like papaya. It was molecularly by Internal Transcribed Spacer barcode confirmed as ABB genotype. It is tolerant to drought and banana bunchy top virus, but susceptible to wilts. Leaf anatomy has three palisades and isodiametric polygonal epidermals. Leaf stomata is amphistomatic, tetracytic on adaxial,and paracytic on abaxial. Pollen unit is monad, very large, subspheroidal, monocolpate aperture, granulum ornamentation, and low viability. Fruit moderately sweet with nutrient contents equal to dessert banana. Pisang Kates has potential as one serving consumer size fruit, ornamental fruit plant, female parent for breeding,and pioneer plant for rehabilitation programs. Both ex-situ and on-farm conservation strategies of this valuable local banana genetic resource are encouraged.

Barangan Merah is a potential type of banana to be developed under in vitro condition. Acclimatization is the final stage in the propagation by in vitro culture. The use of acclimatization medium from organic materials in the surrounding environment can be an alternative in acclimatization process. This study aims to obtain the optimal organic medium in acclimatization process for the growth of Barangan Merah banana seedlings resulting from in vitro culture. The planting material used plantlets from in vitro regeneration of banana cv. Barangan Merah from Tissue Culture Laboratory, Agricultural Faculty, Universitas Syiah Kuala. The experiment used the individual pot technique with the application of organic medium types, namely husk charcoal, sand, leaf litter compost, and oil palm fruit bunches compost. The results of this research showed that the highest number of seedlings that were still survive until the end of the acclimatization process was found in the husk charcoal medium (71.43%). The best seedling growth according to the increasing of leaves number, leaf size, and plant height in the acclimatization process was found in leaf litter compost medium.

Cultivar Barangan is one of the famous banana plants that has good taste and price. The disadvantages of this banana is susceptible to Fusarium disease. Tissue culture techniques is one ways of plants propagation for this plant. This study is aimed to determine the effective concentration of benzyl amino purines (BAP) and indole-3 acetic acid (IAA) for optimal multiple bud clumps (MBC) induction in explants of Barangan Merah banana. The Research was carried out at the Laboratory of Tissue Culture, Faculty of Agriculture, Universitas Syiah Kuala. The research design was a completely randomized design, 3x2 factorial. BAP concentration was the first factor, i.e. 5, 10 and 15 mg L−1. The second factor was the concentration of IAA, i.e. 0.1 and 0.5 mg L−1. The results of this research showed that 96,29% explants of all treatments survived. Explant contamination was 11,11% occurred by bacteria and brownish exudate that spread to the medium. MBC are significantly increased at 16 days after induction at concentration of BAP 5 mg L−1 + IAA 0.5 mg L−1. All combinations of BAP and IAA concentrations have produced MBC explants (100%) at eighth weeks after induction. The concentration of 5 mg L−1 BAP + 0.5 mg L−1 IAA resulted the highest shoots number. Analysis of variance and Least Significant Different further test showed that the longest of shoot length was founding the concentration of 5 mg L−1 BAP compared to other treatments. Increasing concentrations of BAP 10 mg L−1and 15 mg L−1 resulted in significantly lower shoot length. It can be concluded that taken together the concentration of 5 mg L−1 BAP + 0.5 mg L−1 IAA is the best to effectively multiple bud clumps induction in explants of Barangan Merah banana through tissue culture technique.
