Generation of Dipeptidyl Peptidase-IV-Inhibiting Peptides fromβ-Lactoglobulin Secreted byLactococcus lactis

Abstract: Previous studies showed that hydrolysates of β-lactoglobulin (BLG) prepared using gastrointestinal proteases strongly inhibit dipeptidyl peptidase-IV (DPP-IV) activity in vitro. In this study, we developed a BLG-secreting Lactococcus lactis strain as a delivery vehicle and in situ expression system. Interestingly, trypsin-digested recombinant BLG from L. lactis inhibited DPP-IV activity, suggesting that BLG-secreting L. lactis may be useful in the treatment of type 2 diabetes mellitus.

“…Kefir supplementation 3.6 ml/200 g decreased expression of CD26 after exposure to CPF dose 5 mg/kg but not significant. The role of kefir is to inhibit the activity of CD26 through β-lactoglobuline which is produced by Lactococcus lactis and through the ability to decrease the level of IL-2 [46,47]. In this study, the ability of kefir in decreasing expression of CD26 is correlated with the ability to decrease the levels of TNF-α and IL-β, which increased after exposure to CPF and maintain TGF-β level not to decrease.…”

“…The ODN sequences used in this study are shown in Table . The following antigens (AG) were used: Buckwheat (BW, Fagopyrum esculentum , lyophilized form; GREER Laboratories, Lenoir, NC, USA) (Shigemori et al ), α‐casein from bovine milk, ≥70% α S ‐casein (CAS, Sigma, St Louis, MO, USA) (Shigemori et al ), β‐lactoglobulin (BLG, Sigma) (Shigemori et al ), and albumin from chicken egg white (Ovalbumin, OVA, grade V; Sigma).…”

“…Gene expression by gmNZ9000 was induced in 2-or 50-mL scale cultures. The experimental procedures were performed according to previously described methods (Shigemori et al, 2013(Shigemori et al, , 2014. Briefly, an overnight culture of gmNZ9000 was inoculated at 1/20 dilution into fresh GM17cm broth and incubated at 30°C until the culture reached an optical density at 600 nm (OD 600 ) of 0.4.…”
