Fluorescence-based surface magnifying chromoendoscopy and optical coherence tomography endoscope

Abstract: Abstract. A side-viewing, 2.3-mm diameter, surface magnifying chromoendoscopy-optical coherence tomography (SMC-OCT) endoscope has been designed for simultaneous, nondestructive surface fluorescence visualization and cross-sectional imaging. We apply this endoscope to in vivo examination of the mouse colon. A 30,000 element fiber bundle is combined with single mode fibers, for SMC and OCT imaging, respectively. The distal optics consist of a gradient-index lens and spacer to provide a 1× magnification at a wor… Show more

“…OCT can be used to image human colonic crypts , and while our system has the capability to resolve mouse colonic crypts, the limited contrast of the very small crypts in the mouse precludes reliable imaging with OCT. Therefore, SMC is a complementary modality as it can visualize these structures with high resolution and contrast .…”

“…Side-viewing needle probes with diameters of a few hundred microns have also been demonstrated with OCT only 28 and using a dual-clad fiber for OCT and single-point fluorescence detection. 29 Other endoscopes of a few millimeters in diameter have combined various implementations of fluorescence imaging with OCT. [30][31][32][33] None of these examples combines widefield fluorescence imaging and OCT into a submillimeter diameter package. Furthermore, none of these examples includes UV excitation to use MFI for wide-field autofluorescence detection.…”

“…The current imaging modalities that exist with these properties are optical coherence tomography (OCT), reflectance confocal microscopy, and fluorescence spectroscopy and fluorescence confocal microscopy. OCT has the ability to resolve spatial structure with high resolutions of 2-20 μm at depths of 1-2 mm using backscattered near infrared light 25 . Fluorescence spectroscopy and fluorescence confocal microscopy systems provide information about biochemistry used for early cancer detection with depth weighted average fluorescence 69 .…”
