Expression of TGF-β1, SNAI1 and MMP-9 is associated with lymph node metastasis in papillary thyroid carcinoma

Abstract: TGF-β1, SNAI1 and MMP-9 are implicated in tumor invasion and metastasis. The purpose of this study was to examine TGF-β1, SNAI1 and MMP-9 expression in papillary thyroid carcinoma (PTC), and to assess association of TGF-β1, SNAI1 and MMP-9 expression with several clinicopathological indicators of PTC. TGF-β1, SNAI1 and MMP-9 protein expression in 83 PTCs and their matched normal thyroid specimens were analyzed using immunohistochemistry. The mRNA expression levels of TGF-β1, SNAI1 and MMP-9 in 12 fresh PTC spe… Show more

“…Finally, the results confirmed a higher expression of MMP-13 by metastatic cell lines compared to the primary cell line. The latter finding is consistent with studies which showed that MMP-13 expression was significantly increased in tumors from lymph node metastases in head and neck squamous cell carcinomas and papillary thyroid carcinomas patients [11,24] .…”

“…Numerous findings presented in the literature demonstrated a role for MMP-13 in cell migration. MMP-13 silencing was shown to decrease tumor cell migration, whereas over-expression of MMP-13 increased cell migration and promoted metastasis [8,24]. Whether such a role of MMP-13 also counts for the cell types we have used, needs additional experiments.…”

“…It has been suggested that EGFR is highly expressed in aggressive PTC with lymph node metastasis and plays a role in the progression of TC [37], but these results are in contrast with the findings of other studies [38].…”

“…Interestingly, 15 mg/L of the PTF significantly inhibited ERK activation and NCI-N1703 cell migration and invasion, but its reduction of MMP-2 and MMP-9 expression was not significant (P=0.058 and 0.061, respectively). Accordingly, we speculated that other molecular targets, such as other MMPs, may be involved in this antimetastasis mechanism that requires further investigation [29][30][31] . Taken together, the significant findings of this study include the following: the PTF may induce tumor cell apoptosis by regulating the ERK1/2 and JNK MAPK signaling pathway.…”
