Expression of a myeloid marker on TdT-positive acute lymphocytic leukemic cells: evidence by double-fluorescence staining

Abstract: The expression of a myeloid-specific antigen was detected on TdT- positive blast cell populations in two cases of childhood acute lymphocytic leukemia. Double-fluorescence staining by using the monoclonal antibody, VIM-D5, which is specific for cells of myeloid origin, in combination with TdT antiserum revealed that a distinct portion of the blast cells carried both markers. The finding represents the first direct demonstration of this specific biphenotype in leukemic cells and was interpreted as the abnormal … Show more

“…Pui has demonstrated the presence of CALLA in 3 cases of peroxidase positive acute leukaemia (23). Alternatively, the expression of a myeloid marker has been demonstrated in 2 cases of ALL (24). In these rare cases, the coexistence of lymphoid and myeloid markers on the same cells could be explained by the existence of a common progenitor cell which could retain both phenotypes during leukaemic transformation.…”

“…It should be noted that Bettelheim and co-workers have also described two cases of acute leukaemia in which overlapping staining for TdT and a granulocyte-associated antigen, detected by Mab VIM-D5, was observed [29]. However, both appeared to be cases of childhood ALL.…”

“…The finding that colony cells of patient 4 expressed the common thymocyte related antigen T6 was unexpected. Earlier, the expression of myeloid markers on acute lymphoblastic leukaemia cells has been reported (Bettelheim et al, 1982). It was suggested that this phenomenon may reflect derepression of gene activity.…”
