We aimed to study hematological responses of Oreochromis niloticus experimentally exposed to the contaminated water of the Santos-São Vicente Estuary, testing hypotheses that exposure time to estuarine water promotes deleterious effects on hematological parameters and evaluating the use of erythrocytes and leukocytes alterations as environmental biomarkers. Estuarine water was collected from Largo da Pompeba. For the biological assay, 28 juveniles of O. niloticus (red strain) of both genders were randomly selected from commercial pisciculture. For the biological assay, 28 juveniles of O. niloticus of both sexes were randomly selected from commercial fish farms. The juveniles were kept in estuarine water for 72 and 120 hours and, after exposure, blood was collected by puncture of the caudal vein to determine total erythrocytes, hemoglobin concentration, hematocrit, hematimetric indices and total leukocytes, as lymphocyte, neutrophils, monocytes, eosinophils, and basophils were quantified by blood extensions. To test exposure overtime on hematological variables, we performed a two-factor Multivariate Analysis of Variance. Exposure for 72 hours resulted in immunosuppression as seen by the reduced counts of neutrophils, monocytes, and lymphocytes in the bloodstream, whereas after 120 hours the immune system was stimulated with the increase of all leukocyte cell types. Exposure to estuarine water resulted in marked changes in the leukocyte count of O. niloticus, demonstrating that alterations in white blood cells might be more sensitive biomarkers than red blood parameters.

During the burning of the organic material (firewood) in the drying, combustion may occur incompletely, thus generating smoke contaminants such as polycyclic aromatic hydrocarbons (PAHs) in the furnaces. This study aimed to identify possible contamination by PAHs in soybean grains from storage units in municipalities in the Midwest region that have undergone drying in a direct-fired furnace with firewood as fuel. The soybean grains were collected in different municipalities of the Midwest region of Brazil, totaling 22 samples. A survey of possible contamination of soybean grains by PAHs was carried out using High-Performance Liquid Chromatography. The experiment was carried out in a completely randomized design with three replicates, and the data were subjected to analysis of variance, with means compared by Tukey test. The sum of the groups was: PAH4 -1.45 μg kg À1 for Ed eia (2), PAH8 -2.97 μg kg À1 for Catalão (19) and PAHT -5.06 μg kg À1 for Ed eia (2); for benzo(a)pyrene, the sum was below 2.0 μg kg À1 , not exceeded by the value of 0.64 μg kg À1 for Ed eia (1). The values found of PAHs did not exceed the maximum limits established by the European Union, except for infant food.

Bioavailability of polycyclic aromatic hydrocarbons in Santos Bay (Brazil) and its adjacent continental shelf

This study evaluated the bioavailability of polycyclic aromatic hydrocarbons (PAHs) in Santos Bay (SB) and the adjacent Santos Continental Shelf (SCS) in Brazil. Biliary metabolites were measured in several fish species to establish a baseline for future monitoring programs. Bile samples from different species of fish were collected monthly from July to December 2005 in SB, and in August 2005 and February 2006 on SCS. Metabolite concentrations were determined using high-performance liquid chromatography with fluorescence detectors. Naphthalene, phenanthrene, and benzo[a]pyrene metabolite concentrations ranged from 24 to 810 µg g -1 of bile, 1.8 to 68 µg g -1 of bile, and below the limit of quantitation to 1.3 µg g -1 of bile, respectively. Despite its high concentrations, the levels of naphthalene metabolites were in regions of low-contamination, while benzo[a]pyrene metabolite were in the same range as those reported in moderately contaminated areas, which may indicate pyrolytic contamination by PAHs. No significant differences in the metabolite concentrations were found between the SB and the SCS samples or during the periods of collection. Future studies with a single biomonitoring species should be conducted, considering age, sex, and feeding condition of the individuals. The metabolite data presented in this study is an important baseline information for this urbanized region, which hosts several sources of contaminants.
